| Research backgroundIn recent years,as the problem of population aging becomes increasingly serious in our country,how to delay aging,enhance body disease resistance,improve life quality of the elderly and postpone the progress of population aging,has become one of researching hotspots currently.Aging,is an integrated response of changes and hypofunction in the organization structure of multiple organs and systems of life process along with the age growth,is regulated by multi-gene,influenced by multiple factor such as environment and psychology.To delay aging is still potential even though it is an inevitable trend.There have been various theories and opinions on the mechanism about aging,among which immunosenescence has aroused wide attention.Preliminary studies has indicated that there wrer similar effects between Radix Hedysari and Radix Astragali,in the improvement of immune function and antioxidant of aging,especially that the Radix Hedysari was more effective in enhancing proliferation of lymphocyte and secretion of IL-2;there was similar efficacy of immunological enhancement between Yupingfeng Powder containing Radix Hedysari and Yupingfeng Powder containing Radix Astragali,especially that the compound containing Radix Hedysari was more effective in improving the spleen index,enhancing splenic T lymphocyte proliferation and reducing senescent cells.ObjectiveBased on early compared studies on the immune function of single herb Radix Hedysari or Radix Astragali and their compound,this study took the same dose of Radix Hedysari substituting Radix Astragali in Yupingfeng Powder,observed the impacts of Yupingfeng Powder containing Radix Hedysari or Radix Astragali on activation of splenic T lymphocyte in SAMP8 mice by animal experiments,to further elaborate differences in the mechanism between Yupingfeng Powder containing Radix Hedysari and the original compound about delaying aging in the respect of cellular immunity,also supple theoretical basis of the mechanism of anti-immunosenescence for compounds containing Radix Hedysari.MethodsTen Kunming mice(token as young group),forty SAMP8 mice(randomly divided into four groups including Yupingfeng Powder containing Radix Hedysari group,Yupingfeng Powder containing Radix Astragali group,Thymosin group and Blank model group),among which the TCM groups and Thymosin group were respectively administrated at the dose of 6 g·kg-1·d-1 and 4.6 mg·kg-1·d-1,both the Blank model group and Young group were gavaged with normal saline of the same volum.After fourteen days of administration,draw blood by eyeball enucleation to collect serum,then isolate the spleen by aseptic operation to prepare T lymphocyte cell suspension.Detect content of serum cytokines IL-2,IFN-y by ELISA;detect the expression of splenic T lymphocyte surface molecules CD3,CD4,CD8,CD28 and CD 152 by by flow cytometry;detect the expression of it’s CD28 mRNA,PI3K mRNA and p38MAPK mRNA by quantitative real-time PCR;detect the protein expression of it’s PI3K and p38MAPK by Western Blot;detect the protein expression of p38MAPK in the spleen by immunohisto-chemical staining.ResultsCompared with Blank model group,the level of serum cytokines IL-2 and IFN-y increased in SAMP8 mice administrated(P<0.05),specially that the level of IL-2 in Hedysari group was significant than the Astragali group(P<0.05),and the effect of TCM groups were inferior to Thymosin group(P<0.05);the expression of splenic T lymphocyte surface molecule CD8 increased in TCM groups(P<0.05),whereas there had no effect in the Thymosin group,and the expression of surface molecules CD4,CD28CD152 and CD4/CD8 all decreased in administrated groups(P<0.05);the expression of splenic T lymphocyte CD28 mRNA,PI3K mRNA and p38MAPK mRNA all increased in administrated groups(P<0.05),especially that the effect of the Yupingfeng Powder containing Radix Hedysari group was significant than the Astragali group(P<0.05),and except that the effect of Hedysari group and Thymosin group in rising the PIDK mRNA expression are equivalent(P<0.05),other effects of TCM groups were inferior to Thymosin group(P<0.05);The protein expression of splenic T lymphocyte PI3K and p38MAPK all increased in administrated groups as well as the positive rate of p38MAPK(P<0.05),and the effect of TCM groups were inferior to Thymosin group(P<0.05).ConclusionThe Yupinfeng powder containing Radix Hedysari or Radix Astragali both had effects in improving the immune function of SAMP8 mice,especially that the compound containing Radix Hedysari was more effective in enhancing the secretion of serum IL-2 and the expression of splenic T lymphocyte CD28 mRNA,PI3K mRNA and p38MAPK mRNA. |
