Comus officinalis is one of the rare and traditional replenishing yin to tonify kidney and resistance senescence medicinal materials in Chinese medicinal clinic, which is often used after removal of the fruit stone, or wine-baked for therapeutic requirements. The crude has the effects of inducing astringent and arresting sweating, while the wine-baked has better effect of tonifying the kidney and liver, so it is often used in tonics. This paper comparing the herb’s pharmacology before and after processing, and study the active parts of synogism after processing of Comus officinalis. The contents as follow:Part one: study on the resources distribution, cropping, processing, different processing methods of dynasty and today, pharmacologic action and chemical constituent by reading literature, to seek the intention of processing.Part two: study on phamacological effect in pre- and post-processed of Comus officinalis, taking the effect on the nonspecific immunologic function of normal and immunodepressed small rats, the specific humoral immunologic function and the specific cell immunologic function, the small rats with Kidney-Yin Deficiency function, the anti-inflammation and the Relieving cough of small rats, protective effects on the small rats with acute liver injury and the platelet aggregation of domestic rabbits as the index. The results indicate that processing can enhance the effect on the nonspecific and specific humoral immunologic function of immunodepressed small rats, the small rats with Kidney-Yin Deficiency function, protective effects on the small rats with acute liver injury.Part three: study on the active parts of synergism after processing, taking the same index. The results indicate that the petroleum ether part can enhance the effect on the nonspecific immunologic function of immunodepressed small rats, the CHCl2 part can enhance the protective effects on the small rats with acute liver injury, the polysaccharide by ethanol precipitating part can enhance the effect on the specific humoral immunologic function of immunodepressed small rats, the effect of tonifying the kidney and liver and antioxidation.Part four: study the pharmacologic action of the petroleum ether part, taking the effect on the specific humoral immunologic function, the small rats with Kidney-Yin deficiency function and the protective effects on the small rats with acute liver injury as the index. The results indicate that it also can enhance the effect on the specific humoral immunologic function, prolong the hypoxia time. It is thus clear that the petroleum ether part is the active part of synergism after processing.Part five: study on the HPLC of CHCl2 part in pre- and post-processed of Comus officinalis, it indicate that the content of 5-hydroxymethylfurfural in the CHCl2 part increasing 2.5 time after processing. And the pharmacodynamics and cytology of 5-hydroxymethylfurfural, taking the effect on the small rats with acute liver injury, the platelet aggregation of domestic rabbits and the protective effects on the liver cell with H2O2 and CCl4 as the index. The results indicate that 5-hydroxymethylfurfural can obviously protect the small rats with acute liver injury and also have dose-effect relationship, inhibiting the platelet aggregation of domestic rabbits and obviously protect effects on the liver cell with H2O2 and CCl4. It is thus clear that 5-hydroxymethylfurfural is the active component of synergism after processing.Part six: study on the extraction technology of the polysaccharide by ethanol precipitating part, taking the single factor and orthogonal experiment, the most extraction technology as follow: extraction method is recirculation extraction, extraction time is 1.5h, the ratio of solid-liquid is 1∶14, extraction time is 2 time, medicinal materials granularity is 60 mesh, ethanol precipitating temperature is room temperature, ethanol precipitating concentration is 80%, ethanol concentration is 95%, ethanol precipitating time is 8h, sample concentration is 1/10. And the most determination condition as follow: vitriol dosage is 7.0mL, phenol dosage is 1.0mL, reaction temperature is 100℃, reaction time is 20min, and comparing the content of polysaccharide in pre- and post-processed of Comus officinalis, crude comus is 6.28%, processed comus is 4.52%.Study on the pharmacological effect in different molecular weight part of polysaccharide in pre- and post-processed of Comus officinalis, taking the specific humoral immunologic function as the index, the result indicate that 40% polysaccharide can enhance the effect on the specific humoral immunologic function. And study on the pharmacological effect by the other index, it also can obviously enhance the effect of tonifying the kidney and liver and antioxidation.. It is thus clear that the 40% polysaccharide ether part is the active part of synergism after processing.
