Green Thorn Fruit Flavonoids And Alkaloids Isolated Antibacterial Activity

ObjectiveTo extract flavonoids and alkaloids from Prinsepia utilis Royle, to isolate flavonoids compounds, and to study on the antibacterial activity of alkaloids, providing theoretical basis for further developments and utilizations of the natural resource.MethodsThe Prinsepia utilis Royle coarse powder is subjected to continuous extractions, filtrations,obatain flavonoids and alkaloids.By thin layer chromatography, silica gel column chromatography, and gel column chromatography, the monomer compounds are separated and collected from the total flavonoids extracted from Prinsepia utilis; then the chemical structures are determined by both the physicochemical properties of these compounds, and spectrum analysis method(MS\1H-NMR\13C-NMR).Conduct a preliminary study on antibacterial activity of total alkaloids of Prinsepia utilis using two fold dilution method, after the determination of the minimum inhibitory concentration of Staphylococcus aureus, Escherichia coli, Shigella and Candida albicans (MIC),and minimum bactericidal concentration (MBC).Results1Nine compounds have been isolated from the Prinsepia utilis Royle alcohol extracts of ethyl acetate fraction, six of which were identified using physical and chemical properties of modern spectroscopy methods, including4flavonoids:eriodictyol, kaempferol, quercetin, catechin;1organic acids:protocatechuic acid;1sterols:β-sitosterol. Among them, eriodictyol and protocatechuic acid were first isolated from this plant.2The minimum inhibitory concentration of total alkaloids of3Prinsepia utilis Royle on Staphylococcus aureus, Escherichia coli, Shigella and Candida albicans (MIC) were:0.08mg/ml,0.625mg/ml,1.25mg/ml,2.5mg/ml; minimum bactericidal concentration (MBC) were:0.16mg/ml,1.25mg/ml,2.5mg/ml,5mg/ml. Conclusion1Alcohol extracts of Flavonoids from Prinsepia utilis Royle from ethyl acetate fraction are mainly flavonoids.2The alkaloids of Prinsepia utilis Royle test bacteria have certain inhibition effect on four kinds of supply, antibacterial activity:Staphylococcus aureus, Escherichia coli, Shigella, Candida albicans, and its MBC was2times of the corresponding MIC.
