Objective:To explore the efect and the action mechanism of Gengnian decoction by observe the exponent of ovary and uterus, the pathologic morphology of ovary and uterus, the ER and ERmRNA in hypothalamus, the Bax and BaxmRNA in ovarian granulosa cells of Perimenopause model rats.Methods:To build natural aging Perimenopause model rats by the way of estrous cycle and metering each layer cells through the exfoliated cervicall cells selecting of46female SD rats on11-14month age. Then divided the30rat randomly into three groups, the Gengnian decoction group, the textbook decoction group, the normal saline group, when the SD rats meeting the perimenopause model rats requirements. While build12female SD rats on4month age in young control group,12female SD rats on18month age in old control group. To kill rats by blooding from femonal artery after the success of gastric perfusion. Useing uterus, ovary, hypothalamus of rats in each group to test indexes detecting. By adopting weighing conversion method, pathological section, Immunohistochemistry and in Situ Hybridization technique to compared the the exponent of ovary and uterus, the pathologic morphology of ovary and uterus, the ER and ERmRNA in hypothalamus, the Bax and BaxmRNA in ovarian granulosa cells in every groupe.Results:1.Compared with normal saline group:①The difference of Gengnian decoction group had statistical significance on the exponent of uterus(P<0.05), the difference had no statistical significance on exponent of ovary(P>0.05),②In the comparison of pathological section, The difference of follicular development, follicular atresia and follicular expansion which judged the developmental condition of follicular had statistical significance or statistical significance obviously (P<0.05or <0.01). The difference of endometrium, endometrial gland and endometrial stromal which judged endometrium atrophy had no statistical significance (P>0.05).③The difference of Positive Expression of ER in hypothalamus, Bax and BaxmRNA in ovarian granulosa cells of Gengnian decoction group all had statistical significance (P<0.05). 2.Compared with textbook decoction group:①The difference of Gengnian decoction group had no statistical significance on the exponent of uterus and ovary(P>0.05).②In the comparison of pathological section, The difference of follicular development had statistical significance obviously (P<0.01). The difference of follicular atresia had statistical significance (P<0.05). The difference of follicular expansion had no statistical significance (P>0.05).②In the comparison of pathological observation on endometrium and ER in hypothalamus’, there had no statistical significance (P>0.05). The difference of Positive Expression of Bax and BaxmRNA in ovarian granulosa cells of Gengnian decoction group had statistical significance (P<0.05).Conclusion:Gengnian decoction can improving the Perimenopause Model Rats’ exponent of uterus, promoting follicular development, up-regulating the ER in hypothalamus, inhibiting the positive expression of Bax and BaxmRNA in ovarian granulosa cells.
