Objective:Clinical research:The essay studies the therapeutical ly effect of the decoction of geng nian soup on peripheral menopause syndrome (PMS)with people of kidney-yang deficiency or kidney yin-yang deficiency to explore its action and mechanism and treatment of this disease.Experimental research:The essay discussed the effect and the action mechanism of Gengnian soup by watching the pathological tissue of paranephros and hypothalamus, the pathologic morphology of paranephros and hypothalamus, the Bax and Bcl-2 in paranephros and the ER and ERmRNA in hypothalamus of perimenopause model rats of kidney -yang deficiency. Methods:Clinical research:This study observed 60 cases of kidney-yang deficiency or kidney yin-yang deficiency type PMS patient and randomly divided them into treatment group (oral the decoction of geng nian group)and the control group (oral the decoction of Books Group). Then the clinical efficacy of two groups of patients after treatment and E2?FSH?LH, the change of bone mineral density were observed.Experimental research:The study chose perimenopause model rats among female SD rats and divided the rats randomly into four groups, the decoction of geng nian soup, the textbook group, the Livial group and the normal saline group. At the same time, the study chose four-month-old female SD rats and eighteen-month-old female SD rats as compared group. The paranephros and hypothalamus of rats in each group were tested experimentally after killing rats. After that, the paranephros and hypothalamus in each group were compared and the ER and ERmRNA in hypothalamus, the Bax and Bcl-2 in paranephros were tested by using the pathological slices,Immunohistochemical method and Hybridization detection method. Results:Clinical research:The clinical effect of treatment group and compared group are 56.6% and 40%, and there are some differences between two groups. The curative effect of traditional Chinese medicine were 53.3% and 33.3%. The total effect of these two groups are 83.3%?60%.there are also some differences between two groups.(P <0.05)FSH,LH in the blood was lowered, while the concentration of E2 in the blood is raised (P<0.05),The measurement of bone density T quantity was slightly raised after treatment(P>0.05).Experimental research:Compared with normal salin group:?There are statistical significances in zona glomerulosa?zona reticularis and medullary in gengnian soup(P<0.05, P<0.01); there are no statistical significances in zona fasciculata(P>0.05); there are statistical significances in bone marrow stromal and zona reticularis compared with Bcl-2 (P<0.05),there are no statistical significances in zona glomerulosa(P>0.05);?There are statistical significances of ER in hypothalamus;?There are statistical significances of bone mineral density in gengnian soup. Compared with textbook decoction group?there are statistical significance in zona glomerulosa?zona fasciculata and medullary in geng nian soup(P<0.05;P<0.01);?There are ststistical significance of ER in hypothalamus(P<0.05);?There are statistical significance in BMD(P<0.05) in geng nian soup. Conclusion:Clinical research:It is effective to treat the deficiency of kidney-yang or kidney yin-yang deficiency for pre-and post-menopausal women by traditional Chinese medicine from the respects of the bone density, relieve symptoms and FSH?LH?E2. The study has provided subjective materials for the treatments of examples of pre-and post-menopausal women.Experimental research:The decoction of geng nian group can raise the ER in hypothalamus in model rats which are kidney-yang expression of paranephros Bax,increase Bcl-2 in paranephros and strengthen the bone mineral density.
