| Geographical origin traceability is the key to determine the genuineness of herb. Currently herbal origin discrimination methods include morphological identification, physical identification, chemical identification and fingerprint analysis of inorganic elements, but their applicability are limited and less than convincing. The herbs of preparation of traditional Chinese medicine can not determine the origin.In recent years, the application of elemental analysis isotope method to discriminate origin especially in the wine, coffee, juice, beef, mutton has made significant progress. The technology is based on the basis of geochemical information, so this method is used universally. But the application of element analysis on the TCM is blank. Origin is the authentic characterization of Chinese herbal medicine; therefore exterminating herbal origin is one of the key to studying TCM. American ginseng, also known as ginseng, is native to the northern United States to southern Canada. Since ginseng origin is definite and representative. Therefore, this study used ginseng as an example. Domestic and imported ginseng prices are different, so tracing ginseng origin also has practical significance.Ginseng and its preparations were determined by ICP-MS and IRMS.Then we used data compression method, and filter discriminative sense elements to establish the origin discriminant model. The main research contents are as follows:1. Investigation of ginseng source1.1This part lists climate, latitude, longitude, altitude of main origin of American ginseng. In conclusion, sunshine and rainfall and so on are not same.1.2The origins of American ginseng are Shandong (Wenden, Laizhou), Beijing (Huairou), Jilin (Huangni, Fushun, nine), Canada (Vancouver, Ontario), United States (Wisconsin). We totally collected47Samples from those origins. By using inorganic elements we established the model to discriminate the origin.2. Optimization of microwave digesting conditions2.1Digestion system and digestion solution ratio were investigated in this part. The optimum digesting condition is:4:1(HN03and H2O2), warm temperature190℃, holding time10min sample digestion.2.2Ginseng powder was precisely weighed, each about O.1g. We used ICP-MS to determinate inorganic elements of ginseng,concluding Be, Mg, Al, K, Ca, Sc,V, Cr, Mn, Fe, Co, Zn, Ni, Cu, Ge,Se,Ag,In,Sn,Tb, Sr, Bi, Sb, Ba, Hg, T1, Pb, Th, Cd, As, Mo, U. We find out some meaningful elements, such as V, Cr, Mn, Fe, Co, Zn, Ni, Cu. was83.8%of Beijing,77.8%of Jilin,90.9%of Canada and87.5%of Shandong3. Establish the origin discriminant model by isotopeGinseng powder were precisely weighed, each about0.01g, we used IRMS to determine C, H, O, N isotope ratios. We found that the H,O,N were meaningful elements. T The correct rate was72.7%of Huairou,100%of Jilin,100%of Wisconsin USA,70%of Canada and56.2%of Shandong.4. Establish the origin discriminant model of ginseng preparationWe use ginseng powder,excipients,tableting analog to make ginseng tablets, then tablets were dissolved in water and centrifuged. The supernatant was removed, the above operation is repeated three times. Dried ginseng powder was weighed about0.01g. We used IRMS to determine C, H, O, N isotope ratio. Processing of data, there is no comparison before and after. So isotope ratio discriminant model can be applied to discriminate American ginseng tablet. |
