Tea polysaccharides are glycoprotein compounds with biological activity. This study combined the membrane integration technique using microfiltration and ultra filtration separation methods to purify the crude tea polysaccharide solution and then analyze the structure and the activity of the polysaccharide purified, and optimized the technology of buccal tablets of polysaccharides which is extracted from tea. It provides an experimental and theory foundation for a further application of tea polysaccharide. The main research content as follow:(1)This study combined the membrane integration technique using microfiltration and ultra filtration separation methods to purify the crude tea polysaccharide solution and then analyze the structure and the activity of the polysaccharide purified. Results showed that the membrane pore size 0.05μm,0.2MPa operating pressure, PH8.0, and 1% concentration of the material were the most suitable microfiltration process condition, followed by 30K.D ultra filtration membrane secondary purification. The content of polysaccharides processed by this method had been greatly improved, from 50% to 81%. This process condition was simple and suitable for industrial production.(2) TPS was characterized by infrared spectroscopy, high performance gel permeation chromatography, and gas chromatography. Tea polysaccharide was a glycoprotein compound with molecular weight 86994 and 43173. The results showed that TPS was consisted of rhamnose, arabinose, xylose, mannose, glucose, galactose, with mole ratio of 0.63:0.44:0.52:0.48:1.33:0.39.(3) In vitro hypoglycemic experiments showed that:TPS alonged with purification still has a certain extent on α-amylase and α- glucosidase inhibition. The experiment proved the use of membrane separation and purification method was suitable for the separation of heat-sensitive biological products, saved the tea polysaccharide biological activity.(4) Orthogonal design was used to optimize the composition of TPS Tablets that were prepared by selecting malmose as the filler,l%PVP alcohol solution as the adhesive agent, aspartame as the sweeteners and menthol as the freshener. TPS Tablets which prepared according to the optimization method conformed to the Chinese Pharmacopoeia requirement.
