Camelina [Camelina sativa (L). Crantz,popular names gold-of-pleasure,false flax]is anancient crop of the Brassicaceae family.The main objective of this experiment is to cultivate salttolerant and high seed oil content of flax varieties.In previous of this work,we use of60Co-γ raysand"Shenzhou VIII" spacecraft space flight tests independently created luxuriant mutationbreeding material.These materials are used as mutation breeding material.Useing thismaterial,we screened salt stress lines.Using genetic engineering breeding,we clonedseed-specific promoters and built a double T-DNA vector during this experiment.From theArabidopsis totle RNA, was cloned the AtDGAT1and build a plant seed-specific expressionvector pSB130-AtDGAT. Meanwhile,in order screen salt-tolerant false flax from mutationbreeding material,we use MS solid medium as a substrate to200mM of NaCl stressconditions,which screened out of false flax with salt plants.The main results of this study are:1.Cloned by RT-PCR method from the Arabidopsis DGAT1gene to the gene fragmentlength of1563bp,encoding520amino acid sequence corresponding to.2.Ends of cloned gene in AtDGAT1add BamHⅠ and KpnⅠ restriction sites.The geneticconstruct into a plant expression vector.Plant expression vector is double T-DNA vector pSB130for the skeleton to bean promoter and terminator of the original, as the selection markerhygromycin.3.Useing of Agrobacterium-mediated method successfully transferred AtDGAT1into falseflax. We use double T-DNA in order to delet selection marker.This work for screening high oilfalse flax provides new germplasm material.4Through the"Shenzhou VIII" spaceship aviation and60Co-γ breeding,we createdluxuriant mutation breeding material,the total material reaches6000lines. Currently,thesematerials are screened from the22strains tolerant Camelina sativa from2100lines.
