Plateau pika (Ochotona curzniae) and plateau zokor (Myospalax baileyi) are thespecialized native species of Qinghai-Tibet plateau, plateau zokor is one of the blindsubterranean mole rats, and plateau pika is a sprint running animal aboveground. Thispaper had explored the effects of the hypoxic environments on the constitution ofVEGFxxxsplice variants, mRNA levels of the splice variants and the microvesseldensity (MVD) in liver, lung and skeleton muscle of the zokor and pika by moleculartechnology and immunohistochemistry methods. The results indicated that VEGFxxxsplice variants manifestations of differences exist in plateau pika liver, lung andskeletal muscle tissue at different altitude. With increasing altitude, VEGF121hardlyexpressed in plateau pika liver, lung and skeletal muscle tissue. With increasingaltitude which have emerged microvessel density increasing trend, which appears inlung tissue marked increased (P<0.05) skeletal muscle tissue appeared significantlyincreased (P<0.01). These results have prompted VEGF121may play an importantrole in promoting the growth of microvessels. With increasing altitude, there are nosignificant differences VEGFxxxsplice variants in mRNA expression levels in plateauzokor and pika liver and skeletal muscle tissue, but significantly increasedmicrovessel density. These results have prompted VEGFxxxbreduced expression dueto hypoxia and its anti-angiogenic effect is weakened, leading to zokor and pika liver,skeletal muscle microvessel increased. With increasing altitude, plateau zokor lungtissue VEGFxxxsplice variants and its mRNA expression levels and microvesseldensity were marked increased, furthermore, whether high or low altitude, VEGFxxxmRNA expression levels in plateau zokor liver, lung, skeletal muscle tissue weremarked higher than plateau pika, microvessel density is also higher than the plateaupika, which shows that hypoxia can promote the expression VEGFxxxsplice variants,and VEGF can promote microvessel density.
