The study takes Camellia japonica as test material, making a systematic research on thepreservative formula, its anti-aging mechanism and low-temperature storage of Camellia japonicafresh cut flower. On the basis of macroscopical observation of biological characteristics ofCamellia japonica, and through single factor experiment and fine tuning of the orthogonalexperiment on different concentration gradient of preservative ingredient, the study explored thebest preservative formula of Camellia japonica fresh cut flower. After confirming the biochemicalindexes which have close relationship with the senescence of Camellia japonica fresh cut flower,the study then measured the change of these biochemical indexes when the cut flowers had beentreated by preservative. In this way, the study analyzed the mechanism of the preservative’s effecton the senescence of Camellia japonica cut flower. Through studying the vase life and max flowerdiameter of Camellia postharvest cut flowers at different cold storage temperatures and in differentcold days, the study discovered the best cold storage condition for Camellia japonica cut flower.The research results will provide scientific experiment basis and technical support for better use ofthe preservative for the preservation of Camellia fresh cut flowers, as well as for the establishmentof matching techniques for fresh-keeping postharvest cut flowers of Camellia. The major researchresults of the study were as follows:1. Research of the preservative formula for Camellia japonica. As the sugar is the mainnutrition and energy sources for the whole physiological and biochemical process, the first stepwas the tolerance experiment of sucrose concentration, to preliminarily screen out20g/L sucrose asthe best carbon source. Then the study set10kinds of preservative ingredients, with eachingredient having three concentration gradients, for single factor experiment study. Thepreservative ingredients with the same physiological effects were set with two reagents: fungicide(8-HQS and SA), organic acids (CA and BA), ethylene inhibitor (STS and CoCl2), plant growthregulator (6-BA and B9), and inorganic salt (KCl and Al2(SO4)3). The experiment selected thepreservative ingredient for Camellia japonica and found the optimum concentration was SA:50mg/L, STS:0.2mM,6-BA:25mg/L, Al2(SO4)3:100mg/L. Through fine-turning theconcentrations of above selected preservative ingredients, the study carried out an orthogonalexperimental design with each optimum concentration set two fluctuations, thus obtaining9kindsof preservative formula. The best preservative formula of cut Camellia was found and determinedby investigating the vase life of fresh cut flowers and measuring the physiological indexes such asmax flower diameter, fresh weight rate and water balance value. The best preservative formula was as follows:20g/L sucrose+50mg/L SA+0.2mM STS+30mg/L6-BA+75mg/L Al2(SO4)3.2. Research of the anti-aging mechanism of preservative for the Camellia japonica cut flowers.The preservative formula obtained from above experimental results was used to observe its effecton postharvest physiological indexes of the Camellia japonica cut flowers, such as soluble sugar,soluble protein, free proline, MDA, POD activity and relative membrane permeability, withdistilled water as a control. The results showed that the preservative had a better effect than thedistilled water. The preservative could effectively slow down the decline of the contents of solublesugar and soluble protein in cut flowers’ petal, inhibit the ascensional range of the contents of freeproline, MDA and relative membrane permeability, and delay the decline of POD activity. Theconclusion was that the preservative obviously had an important role in the anti-aging of Camellia.3. Research of the cold storage condition of Camellia japonica cut flowers. The study setCamellia fresh cut flower for cold storage at different temperatures of0,2,4,6,8℃respectivelyfor5days, and then measured and analyzed its vase life and max flower diameter. The resultsshowed that the best cold storage temperature for the cut flower was4℃. And in the4℃lowtemperature storage experiment, the study measured the physiological indexes in the cold storagedays of0,2,4,6,8days respectively. The result indicated that the vase life and max flowerdiameter of cut flowers showed a trend of gradual decline with the extension of storage time, andthe best cold storage period of Camellia fresh cut flowers was4days.
