One day old chicks fed with probiotics were used in this study. The tissue paraffin section,periodic acid-schiff stain method(PAS), alcian blue-periodic acid-schiff stain (AB-PAS), transmission electron microscope(TEM), enzymelinked immunosorbent assay(IELISA), and real-time fluorescence quantitative PCR were used for discovering the changes of the number of goblet cells in intestinal tract-ileum, cecum and colon, the content of MUC2in intestinal fluid and MUC2mRNA in intestinal tract. The study aimed at revealing the influence of the number of goblet cells and the content mucin-MUC2in intestinal of chicks after the application of probiotics, which to provide experimental evidence for probiotic research, reduce the risk of chicks intestinal diseases. The results showed that:1.Chicks after the application of probiotics, the total numbers and the numbers of acidic, double-stained, neutral goblet cells in ileum, cecum, colon were different degrees higher than those of controls; the total numbers of goblet cell in chicks’ ileum and cecum were both enhanced compared with those of controls at1~4days(P<0.05or P<0.01), the total numbers of goblet cells in chicks’colon were enhanced compared with those of controls at1~7days (P<0.05or P<0.01); the numbers of acidic, double-stained goblet cells in chicks’ileum were enhanced compared respectively with those of controls at4days and1~4days (P<0.05); the numbers of double-stained goblet cells in chicks’cecum were enhanced compared with those of controls at1~4days (P<0.05); the numbers of acidic, double-stained goblet cells in chicks’colon were both enhanced compared with those of controls at1~7days (P<0.05or P<0.01), and the numbers of neutral goblet cells were enhanced compared with those of controls at1~4days (P<0.05). Those results indicated that probiotics could promote the number of goblet cells and enhance the immune function of intestinal local mucosa.2.Chicks after the application of probiotics, the content of MUC2in ileum, cecum, colon fluid and the expression of MUC2mRNA in ileum, cecum, colon were different degrees higher than those of controls; the content of MUC2in ileum, colon fluid were enhanced compared respectively with those of controls at1~7days and1days (P<0.05or P<0.01); the expression of MUC2mRNA in ileum, cecum, colon fluid were enhanced compared respectively with those of controls at1~4days,1days and1~7days (P<0.05or P<0.01). Those results indicated that probiotics could modulate the content of MUC2in intestinal fluid, upregulate the expression of MUC2gene, could lubricate and protect the intestinal tract, improve the non-specific immune fuction of intestinal tract.3. Tissue ultrastructure examination revealed that the number of mitochondria, golgi, ribosomal, endoplasmic reticulum and mucin secretion in the cytoplasm of goblet cells were enhanced compared with those of controls. Those results indicated that probiotics could promote the growth of goblet cells organelles and the mucin section of goblet cells.4.Two kinds of probiotics that were used in our experiment, chicks after the application of Lactobacillus, the influence of the number of goblet cells and the cintent of mucin-MUC2in intestinal was better than those after the application of Biofidobacterium.
