This study was about the plant tissue culture of Begonia sanguinea Raddi which tookleaves as explant materials and obtained the complete regenerated plants. This experimentwas carried out around several aspects such as the handling and sterilization of explantmaterials,induction and differentiation of callus,multiplication of adventitious bud andinduction of strong seedling, rooting and transplanting of the plantlets in vitro.With the useof single-factor or two-factor, and multi-level experiment design,this research was focusedon a variety of problems including the selection of sterilizing time of mercuric chloride,concentration and ratio of the plant growth regulator,concentration of activated carbon,type of transplantation medium and so forth,setting up a complete tissue culture and rapidpropagation system, which provided technical support for its exploitation,protection ofgermplasm resources and industrial production.The results show that:(1) It was strict for leaves of Begonia sanguinea Raddi to chose the appropriate timeto be sterilized by0.1%of mercuric chloride.Deficiency of sterilization time could increasethe rate of pollution of explants,and long time influence the survival of explant.The beststerilization method was sterilization for8s with75%alcohol,sterilization for6~7min with0.1%HgCl2.(2) The best medium for induction and differentiation of callus was MS+6-BA0.5~1mg/L+NAA0.1mg/L,which was easily induced callus and adventitious buds.Its inductionrate was85.42%~87.08%.(3) The appropriate medium for multiplication of adventitious bud was MS+6-BA1mg/L+NAA0.1mg/L.Explants of this medium induced abundant buds which was strongand proliferated rapidly with proliferation ratio as high as42.43.(4) Adding a certain concentration of active carbon to the medium was advantageousto the strong seedling,with1%concentration is the best.(5) The optimum medium for rooting of test-tube seedlings was1/2MS+NAA0.3mg/L,in which medium there inducing a large number of strong roots with a high speed.The rooting rate could reach100%in20days.(6) The best transplanting substrate for tissue culture seedling is vermiculite:coconutchaff=1:1,whose transplanting survival rate is as high as96.67%.
