Polygonatum.sibiricum Red.is a perennial herb of the Polygonatum Genus belongs to theLiliaceae family.The rhizome of it usually used as a traditional chinese medicine. ThePharmacopoeia of China (2010) reproduces P.sibiricum Red., P.cyrtonema Hua. andP.kingianum Coll.et Hemsl. as its original plants.There are many wild P.sibiricum Red.distributed in Lueyang of Shanxi Province., the area began the study of wild P.sibiricum Redbecomes home after2000. After ten years of development,the area has formed a set of simplecultivation techniques, but some new problems, such as,the reproduction is not clear, thedegradation of germplasm resource,have become the important restrict factors of production. This study provided a theoretical basis for seed propagation techniques of P.sibiricum Redbased on observation of growth dynamic of the seedlings and exploration of appropriatetransplanting age and determined the suitable harvest age and period through the study ofaccumulation of the active ingredients of perennial wild and cultivated P.sibiricum Red.Themain results are summarized as follows:1. The seedlings propagated by seed can’t emergence of soil the first year,growing a leafseedlings close the ground the second year and three or four leaves the third year. Thegermination time ranges from early May to mid-June and the growth time sustained only twomonths.The aboveground of seedlings began to wilt from August.The diameter,length andfresh weight of the rhizome of seedlings are in rapid growth stage before September and thediameter, length of rhizome changes slowly but the fresh weight do not change afterSeptember. Then roots into hibernation. With the increase of growth year, the content of polysaccharide and diosgenin growing quickly, but overall are at a low level.2. The emergence rate of annual seedlings transplanted to the understory and field wererespectively0and2.7%and is unfit for transplanting.The emergence rate of biennial seedlingstransplanted to field was4.4%, while its reaching46.3%to the understory and is suitable fortransplanting to the understory. We give the suggestion that the seedlings growing more thanthree years can used to be transplanted. The measurement results for each index of thebiennial seedlings transplanted to field and the understory showed that understoryenvironment is more suitable for growing seedlings of P.sibiricum Red.. Daejeontransplanting should strengthen field management, creating a suitable environment forP.sibiricum Red growth.3. The content of polysaccharide and diosgenin of the rhizome of three-year cultivatedP.sibiricum Red reached the highest levels by the end of October, while the six-year rhizome has highest level in the early September. The content of active ingredients of the rhizome ofP.sibiricum Red. are less than six–year rhizome before October,while higher after the October. The polysaccharide content are sequentially lower with the age increasing of differentsections of rhizome of six-year cultivated P.sibiricum Red and the diosgenin content in firstthree age sections is relatively higher than the others. So we give the suggestion that theappropriate harvest period and years of cultivated P.sibiricum Red are the end of October andthree years. The determination of active ingredient content showed that the polysaccharideand diosgenin content in wild P.sibiricum Red are lower than the cultivated P.sibiricum Red.The polysaccharide content in first three age sections is less than7%that formulated byPharmacopoeia indicated that the quality of wild P.sibiricum Red is less than the cultivatedP.sibiricum Red.
