Polygonatum.sibiricum Red., is a plant of the family Liliaceae and perennial herb. Itsrhizome usually used as a traditional staple medicinal material. As ceaseless researches in therule of pharmacology, the demand of Polygonatum.sibiricum’s market increases year by yearwith wide prospect. Therefore manual cultivation of Polygonatum.sibiricum Red. became anew way to meet market demand and increase farmers’ income. Thus it is quite important tostudy the Propagation technology of Polygonatum sibiricum.Different means of Polygonatum sibiricum Red. were selected to investigate the effect ofmediums on seed genermination, the effects of plant growth regulator on survivalrate ofgerminating and growthing. The results were as follows:1. Storing at0℃low-temperature sand under the condition of120days is favorite forPolygonatum sibiricum seed germination; Soak for15minutes with3.0%H2O2disinfectioneffect is best. After soaked18-24hours in35-40℃lukewarm waters is conducive to increasethe germination rate and potential of Polygonatum sibiricum seeds. Seed germination rateunder dark conditions is higher than that under illumination of high germination rate,indicating that dark was beneficial for the germination of Polygonatum sibiricum seeds.2. Suitable concentration of GA3,6-BA, salicylic acid and chitosan increased thegermination of Polygonatum sibiricum seeds significantly, and the germination time shortenedremarkably. The concentration for brassinolide (0.005mg/L) seed soaking24hours had thebest effect, germination rate, germination potential, germination index and vigor index wereup to91.11%。During this experiment,100mg/L6-BA still can break bud dormancy andpromote the growth of the aerial parts of Polygonatum.sibiricum Red., in addition to promoteseed germination.3. Stratifcation treatment, exogenous hormone (GA3、6-BA) and CaCN2has a certainimpact on bud dormancy of Polygonatum sibiricum stems.2.0mg/L6-BA can contribute tothe emergence of Polygonatum sibiricum stems, which is significantly higher than the controlgroup.4. Bud inducement was initiated on a MS medium containing6-BA2.0mg/L,2,4-D3.0mg/L,30g·L-1sucrose and5g·L-1agar, and the pH was adjust to5.8. The bestmedium for induction of stem and leaf callus formed was MS+6-BA2.0mg/L NAA+3.0mg/L.
