Pleurotus nebrodensis, also known as Pleurotus ferulae Lenzi,its nutrition is very rich,its polysaccharide has a variety of health function。Taking Pleurotus nebrodensis strain KN-01 as the material,this paper studied five factors such as the nutrition physiology,light illumination,low temperature stimulation method and ripening management, and optimization of cultivation formula, in order to solve the problems,such as the relatively lower production,the longer production cycle,to provide reference for high-yield cultivation of Pleurotus nebrodensis.The main results are as follows:1. Taking Pleurotus nebrodensis strain KN-01 as the material, its colony diameter as the mian index, and cultivated on the medium added with different type and concentration of components, such as carbon source, nitrogen source,vitamins, inorganic salt and plant growth regulator.The purpose of this study was systemically analyzing the effect of different components on mycelium growth of Pleurotus nebrodensis.As a result,variety of carbon sources can be used by Pleurotus nebrodensis, with soluble starch, mannitol and arabinose as carbon source,the mycelial growth was the best, with substances,carboxymethyl cellulose, maltose and glucose as carbon source, the growth of mycelium was better, while sorbitol and xylose as carbon source, the growth of mycelium was poor.Aspartic acid had strongest promoting effect, leucine and isoleucine followed, colony diameter were respectively increased 174.5%, 101.6% and 21.7%.(NH4) 2SO4, FeC6H5O7.NH4 OH,(NH4) 2HPO4 and MgSO4 had obvious promoting effect on the colony diameter,corresponding most sustable concentration respectively were1.8 g·L-1,1.3 g·L-1, 1.1 g·L-1 and 0.5 g·L-1. Compared with controls, the colony diameter respectively improve 61.1%, 65.3%, 62.4%,62.7%.While KH2PO4 and Al(NO3)3 had a stronger effect on the mycelium growth,and the colony diameter increased by 46.8% and 44.4% respectively.2. The study was to explore the influence of light environmental factors,such as light color, illumination time, illumination intensity, which required for a successful basidiome development,on the growth and dvelepoment of Pleurotus nebrodensis.The conclusion showed that,blue exhibitd advantages of improving yield,which increased by 4.61% than the white light;Under the red light,yellow light and green light, the basidiocarp showed slow development,poor shape and low yield.In addition, in the development stage of fruiting body,light intensity should be 500~700 lx;the most suitable illumination time was 11.2 h·d-13. With the mixture ratio and four factors quadratic general rotary unitized design,industrial cultivation formula of Pleurotus nebrodensis was optimized in this chapter.The conclusion showed adding wheat bran, corn meal, soybean meal to the cottonseed shell can significantly increase the yield of Pleurotus nebrodensis,The proportion of sawdust should be lower,otherwise,the yield will be reduced.The highest yield substrate was preliminarily determined,its was mainly consist of 15.6% wheat bran, 3.7% corn powder,6.8% sawdust,4.1% soybean meal,67.8% cottonseed shell;Among the twenty different formulations assayed,the formula 3 and the formula 7 were the faster, the density of the fruiting body of the formula 6 was the maximum, the diameter and the length of the fruiting body of the 4 formula were the larger.4. The experimental results of different low temperature stimulation method and ripening time on mycelium of Pleurotus nebrodensis test showed that the second method was the best low-temperature stimulation mode,namely,stimulating continuously for 8 days at low temperature of 5±1℃;Moreover, in the test range,the appropriate riping time was 50~60 d.
