Platanus(Platanus acerifolia wild) is a world famous street tree and court shade tree species which has broad application on landscape gardening. However, since sycamore has a serious phenomenon of floating seed hair in spring, it is likely to cause pollution of the urban environment and affect people’s lives. In recently years, the emergence of less fruit less seed hair new sycamore species has largely improved the floating seed hair phenomenon in spring as well as the environment quality of urban area. Tissue culture can rapidly propagate seedlings and maintain the good traits of parent species to the maximum possible extent; which makes it ideal for less fruit less seed hair sycamore seedling breeding. This study researches on the fast tissue culture techniques of a less seed hair sycamore species bred by Wuhan Municipal Research Institute for Landscape Gardening, the "Yuanke 115"(Platanus occidentalis’Yuanke 115’); and strives to provide a solid research basis for rapid breeding and factory production of this less seed hair sycamore species.This study used the stem with bud of’Yuanke 115’sycamore as explants for tissue culture; established infrapetiolar bud tissue culture system for less seed hair sycamore. The main findings are as follows:(1) The best sterilization time for stem section with bud is 30 seconds in 70% ethanol, 0.1% HgCl2 for 8 minutes in spring and summer; 6 minutes in autumn and winter.(2) Young stem section with bud has best inducing effects in MS+0.3mg/L 6-BA+0.01 mg/L NAA, under which condition a 86.67% induction rate is achieved.(3) In primary culture, cryogenic dark processing for 3 days and adding 1.0g/L activated carbon (AC) to the culture medium can significantly reduce the browning phenomenon of sycamore during tissue culture; and is conducive to the germination and growth of axillary buds later.(4) The best proliferation medium for stem section with bud is MS+0.5 mg/L6-BA+0.03 mg/L NAA, attaining a proliferation coefficient of 3.91; the number of subcultures should be controlled at less than 3 times.(5) Supplementing the medium with 20mg/L vitamin B4 (Ad) plays a good role for strengthen seedlings; seedling height becomes significantly higher.(6) Best rooting medium for test-tube plantlet is 1/2 MS+0.05 mg/L IBA+0.03 mg/L NAA+10 g/L sucrose +6.5 g/L agar, obtaining a 93.67% rooting rate.(7) A week of water culture for the rooted seedling before transplanting combined with 1:1:2 mixture base materials of humus, perlite and garden soil for transplanting can significantly increase the transplanting survival rate of rooted seedlings to 82%.
