Sugarcane belongs to the genus Gramineae, is native to and widely cultivated in tropical and subtropical regions for sugar production. And it is the most important crop in Guangxi. In this study, using MSAP detection the level of the methylation 8 sugarcane varieties in the elongation stage and 5 sugarcane varieties in the maturation stage. The mine result as follows:1.Using 25 pairs of primer combinations to amplify. The patterns of DNA methylation show that, the methylation polymorphic fragments at the elongation stage was 4719, divided into 3 kinds of type, the total of non-methylated bands was 4288; the total of the fully methylated sites was 241, the number of hemimethylated sites was 190. Therefore, the main way of the sugarcane genomic in CCGG sites methylation is the internal cytosine (Cm5CGG) double stranded DNA methylation.2.The patterns of DNA methylation show that, the methylation polymorphic fragments at the maturation stage was 3307, divided into 3 kinds of type, the total of non-methylated bands was 3213; the total of the fully methylated sites was 171, the number of hemimethylated sites was 41. Therefore, the main way of the sugarcane genomic in CCGG sites methylation is the internal cytosine (5CmCGG) double stranded DNA methylation.3.The changes of the methylation of sugarcane in elongation stage and mature period was:the average of semi-methylation rate was decreased from 4.12% to 1.16%, methylation sensitive amplified polymorphism decreased from 9.12% to 6.02%.4.All data on the Hpa II enzyme of 8 sugarcane varieties elongation stage in MSAP using UPGMA NTSYSpc software, cluster analysis. Guitang 06-1721 was alone to become the first class; Liucheng 05-136 and Guitang 07-645 became the second class; cited Guitang 04-1001 and Guitang C1-2003 as the third class; Guitang 03-1438, Guitang 05-3626 and Guitang 06-244 for the fourth class. With MspI cut data clustering analysis, Guitang 06-1721 was alone to become the first class; Liucheng 05-136 was alone to become the second class; cited Guangxi C1-2003, Guitang 05-3626, Guitang 03-1438, Guitang 04-1001 and Guitang 06-244 for third class.5.The Hpa Ⅱ enzyme of 5 sugarcane varieties mature period in MSAP cutting data for clustering analysis. Guitang 07-645 alone to become the first class; Liucheng 05-136 became the second category; Guitang 05-3626, Guiting C1-2003 and Guitang 06-244 became the third class; Using MspI enzyme to analyze data for clustering, Liucheng 05-136 was alone to become the first class;Liucheng 05-136 and Guitang 07-645 become the second class; cited Guangxi C1-2003 and Guitang 05-3626 for third class.6.The sequencing results of part of sugarcane methylation polymorphism bands showed that:one gene had a high similarity to saccharum hybrid cultivar R570 clone BAC while the other showed vary high homology with sorghum bicolor hypothetical protein. These genes were likely to be related to the agronomic traits of sugarcane, but their function remained to be further identified.
