Gerbera jamesonii Bolus is an important cut flower in the market. In vitro conservation ofgermplasm resources including those cultivars with good properties are valued greatly on a globle scalecurrently.It is practically vital to explore conservation in vitro of Gerbera. Aseptic seedlings of ‘Ziyan’and‘Heixinhuangguan’ treated by different concentrations of PP333to investigate effects of PP333on invitro conservation of Gerbera in multiplication and rooting process in the paper.The main researchresults are as follows:1. Germplasm resources of Gerbera were collected.The experiment collected sixteen kinds of Gerbera and analyzed characters of flower colour,production and stress resistance. In these varieties, characters of ‘Beijixing’,‘Heixinhuangguan’,‘Ziyan’and‘Jinbianjuhuang’ were higher yielding and hardiness were better than others.2. An aseptic culture technique system for Gerbera was established in the paper.Seven kinds of aseptic seedlings including ‘Jintaiyang’,‘Huanglvxin’,‘Heixinhuangguan’,‘Ziyan’,‘Linlong’,‘Jinbianjuhuang’ and ‘Dahonglvxin’ had successfully been cultured.3. Effects of PP333in multiplication and rooting of Gerbera were studied.Treated with various concentrations of0-2.0mg·L-1PP333in MS media, the results showed that insubculture0.8mg·L-1were the optimum concentrations of PP333for ‘Ziyan’ and ‘Heixinhuangguan’.0.8-1.2mg·L-1PP333could dwarf plantlets, suppress proliferation and be used to conserve Gerbera‘Ziyan’ resouses for medium time.0.8mg·L-1PP333could inhibit multiplication of ‘Heixinhuangguan’,but the plantlets had general growth condition, a handful of bleaching, and after recovery growthregeneration plants grew well.Treated with various concentrations of0-2.0mg·L-1PP333in rooting culture,‘Ziyan’ treated by0-0.1mg·L-1PP333which grew better than others, was not albino and vitrified. The plantlets of‘Heixinhuangguan’ treated by PP333had weak growth and stub roots. Plants treated by0.8mg·L-1PP333grew relatively well, had dark-green leaves and was albino rarely.4. Effects of PP333in vitro conservation of Gerbera were studied.0.8-1.2mg·L-1concentration of PP333could delay growth about120days for conserving ‘Ziyan’,and compared with the general subculture, preservation time was extended by60days. Morphologycharacteristics of plants after recovery growth were the same as the normal plants. Effects of PP333on ‘Heixinhuangguan’ were not good.
