Hypodermatid myiasis is a kind of disease which is caused by the fly larva ofhypodermatidae and hypoderma to infect vertebrate. Larvae parasitizes under the skin ofinfected host, the host who appears skin irritation and skin tissue is damaged such symptoms.At present, in the worldwide it can be found that in the hypodermatidae and hypoderma,animals can be infected by h. bovis, h. lineatum, h. sinense, h. tarandi, h. aecton, h. diana andnewly discovered pallasiomyia boulard, among them, cattle are easier to be infected as well asgoats, antelopes, sheep, deers, horses, mice, rabbits, dogs, orangutans such animals andpeople can be infected, but the number of infection is relatively less. It has brought aboutterrible harm and economic losses in animal husbandry, especially to ruminants. In April2000,in the zoo of Shihezi, a kind of third stage larva of unknown torsalo was found under the backof two dead wild argali which had been caught in the Bole mountainous area, the larvae werecollected and stored. After referring to the relevant literature, the report has not beendiscovered.This study is based on the current morphological observation and molecularclassification method of torsalo larvae to identify. It is aimed at exploring and confirming theXinjiang wild argali to infect the third stage larvae species of torsalo.1、The morphological observation in the third stage larvae of unknown torsalo at Xinjiangwild argali back subcutaneousAccording to take morphological observation for Xinjiang wild argali infected the thirdstage larvae’s false head, somite, somite thorn(is mainly about D-iii back and belly, D-X belly)and stigmal plate, at the same time, comparing with the third stage larvae of h. bovis, h.lineatum, h. sinense, h. tarandi. The result shows that Xinjiang wild argali infected the thirdstage larvae of unknown torsalo, its morphological characteristics are similar to the third stagelarvae of h. lineatum, the main similarities are like these:1、there are two fleshy protrusions infalse head’s dorsal part both Xinjiang wild argali infected the third stage larvae of unknowntorsalo and h. lineatum;2、in the second somite’s dorsal part have3clusters thorns and5clusters thorns in veutro, in the arrangement of thorns are also mostly accordance.3、in thethird somite’s dorsal part, the size of front row’s thorns and the arrangement pattern areextremely resemble, in the middle also have rounded fleshy protrusions;4、there isn’t thorn inthe tenth somite’s venter;5、two sides stigmal plate are symmetrical, spiracular button has alittle protuberance, close to C ring, C ring is not occluded. Individual parts havemorphological differences, there are differences:1、Xinjiang wild argali infected the thirdstage larvae of unknown torsalo, in the third somite’s dorsal part, its thorns arrange like a band, but in the same place of h. lineatum’s third stage larvae, there aren’t thorns.2、Xinjiangwild argali infected the third stage larvae of unknown torsalo, in the third somite’s veutro,there are thorns in the front row, back row isn’t, in the middle of somite, irregular circularprotuberance. But h. lineatum’s third stage larvae, there are thorns both front row and backrow in the third somite’s veutro. There are huge morphological differences between othertorsalos’third stage larvae.2、Partial segment clone and sequence analysis of COI gene in Xinjiang wild argali third stagelarvae of unknown torsalo.According to clone the partial sequences of COI gene in Xinjiang wild argali the thirdstage larvae of unknown torsalo, combining with MAGA5.0and DNASTAR to take sequenceanalysis and compare homology of COI gene sequences. A phylogenetic tree is built. Theresult shows that the homology is very high between the partial sequences of COI gene inXinjiang wild argali the third stage larvae of unknown torsalo and h. lineatum, it reaches99.3%. The homology is relatively low with other torsaloes, they are95.6%,94.1%,93.5%,92.0%,91.8%and90.1%; From the phylogenetic tree, the genetic distance is shortest betweenXinjiang wild argali third stage larvae of unknown torlaso and h. lineatum, and at the samebranch.3、Partial segment clone and sequence analysis of18S rRNA in Xinjiang wild argali thirdstage larvae of unknown torsalo.According to clone the partial sequence of18S rRNA gene in Xinjiang wild argali thirdstage larvae of unknown torsalo, combining with MAGA5.0and DNASTAR to take sequenceanalysis and compare homology of this gene, a phylogenetic tree is built. The result showsthat the homology is highest between the partial sequences of18S rRNA gene in Xinjiangwild argali third stage larvae of unknown torsalo and h. lineatum, it reaches99.6%. Thehomology is relatively low with other torsaloes, they are97.2%,98.4%,97.4%,98.3%,98.1%,98.6%; From the phylogenetic tree, the genetic distance is shortest between Xinjiang wildargali third stage larvae of unknown torsalo and h. lineatum, and at the same branch. Theresults are basically consistent with COI gene analysis.According to analyze and compare the research results, Xinjiang wild argali third stagelarvae of unknown torsalo is close to h. lineatum third stage larvae, it can be confirmedprimarily as h. lineatum subspecies.
