Euonymus koopmannii only distributed in the western mountainous area of Tianshan in Xinjiang of China, mainly being born in mountainous region and forest edge and bushwood. E. koopmannii cut resistance, disease and insect resistance, high ornamental value, being wild ornamental plants in Xinjiang with development value.But in recent years, due to the deterioration of ecological environment and human overgrazing and other reasons, makes it be in endangered edges. Therefore, in order to protect and develop it reasonably, carrying out efficient, fast, timely, stable breeding technology of E. koopmanniihas become an urgent problem to be solved. This paper takes the flowers, seeds and leaves of E. koopmannii as study objects, using observation of floral traits and pollination characteristics; in the breeding system, sexual reproduction, asexual reproduction tests respectively; temperature, light, concentrated sulfuric acid and stratification; different disinfection, basic medium and hormone concentration and other methods to study on the type of breeding system, seed germination conditions and the isolated culture of explant. The main results are as follows:The flower of E. koopmannii is bisexual. Flowong 5~6 months, Fruiting 8~9 months.Pistil 1; the ovules of each flower are 6 to 8; the flower disc is fleshy, placentation axile. Theratio of pollen and ovule(P/O) is about 532~1267, hybrid index(OCI) being 4.Through the flower bagging and pollination experiment we can determine that the breeding system of E. koopmannii is the type of being mainly outcrossing,partially self-compatible, needing pollinator. According to the field observation and pollination experiment we can infer that the plant takes the anemophily as the main method. The difference of the expression of sexual function of male and female is the main reason to influent the self-pollination.The seed of E. koopmannii has the characteristic of seed coat dormancy. The optimal germination temperature and reagent matching treatment of the seed in the condition of constant temperature and dark: processing 5 min with 25°C+concentrated sulfuric acid, the final germination rate being 81.42%. The change of temperature could inhibit the seed germination.The light has negative effectson Seed Germination.Aril inhibites the seed germination of E. koopmannii, the germination rate of the seeds with aril in culture conditions of constant temperature being 0. The seed germination rate is the highest in the process with concentrated sulfuric acid 0 min(Potassium Permanganate disinfection method) in thestratification in the ordinary temperature 25°C, the final germination rate being 89.39%, the late seedling quality being more stable.Taking the leaves as the explant, in the pH5.8 ~ 6 disinfection conditions, the effect of taking process of 0.1% mercuric chloride for 5 min being the best, the pollution rate and browning rate respectively being4.6% and 8.2%. In different basic medium, especiallythe treatment effect of in MS basic medium is the most ideal. In different combination of hormone concentration, the growth of the explant callus in MS+6-BA3.0 mg/L+IBA0.2 mg/L is the best, the inductivity of callus having reached 98%. Taking disinfection treatment by 75% alcohol 0.5 min+0.1% mercuric chloride 5 min, the growth of the inducedcalluschanging over into the culture medium of MS+6-BA3.0 mg/L+IBA0.2 mg/L+30 g sucrose +7 g agarbeing the best and the effect being most significant.
