Favolus squamosus also known as old wood fungus, wide-scale edge volvatus, microlepia big volvatus, wide phosphorus Polypores, etc. Belongs to Basidiomycota, Basidiomycetes, Polyporales, Polyporaceae, Boletus. Its fruiting body are hypertrophy and delicious, Favolus squamosus is a new mushroom development with high value and biological activity. This article we choose Favolus squamosus from Qinghai Province to research. Its liquid fermentation nutrient conditions, fermentation technology, extracellular polysaccharide fermentation technology, fermentation kinetics, and the effect of inhibitory, memory, fatigue resistance, antihypoxic of extracellular polysaccharide was studied. The results are as follows:1 Liquid fermentation nutrition conditions and fermentation process of Favolus squamosus1.1 Research on fermentation impact of biomass and nutrient conditionsThrough single factor and orthogonal experiment, we chose a better fermentation process for collecting ball mycelial biomass. That is nutritional conditions of 3% sucrose concentration, 0.1% beef extract concentration, 0.3% KH2PO4 concentration, 0.15%Mg SO4 concentration. Fermentation process conditions of 120ml/250 ml liquid volume, 27℃, 150r/min, 15 glass beads, 4% Inoculum, p H value of 7, culture 6 days, this process is better, reach to 0.463g/100 ml.1.2 Research on extracellular polysaccharide production and nutrient conditionsThrough single factor and orthogonal experiment, we chose a better fermentation process for collecting ball mycelial biomass, that is nutritional conditions of 3% lactose concentration, 0.1% peptone concentration, 0.3% KH2PO4 concentration, 0.15%Mg SO4 concentration. Fermentation process conditions of 120ml/250 ml liquid volume, 27℃, 160r/min, 15 glass beads, 3% Inoculum, p H value of 8, culture 6 days, this process we got relatively high extracellular polysaccharide production, reach to 0.517g/100 ml.2 Research on Fermentation kinetics of Favolus squamosusThe fermentation kinetics results showed that fermentation liquor absorbance values, extracellular polysaccharide production, biomass and the mycelium ball diameter along with the continuous fermentation time increase, p H value converse. Each value is stable on 7th day, so the end of fermentation is on 7th day.3 Study on antibacterial activity of Favolus squamosus extracellular polysaccharideAntibacterial results showed that Escherichia coli, Bacillus subtilis, Staphylococcus aureus, Monilia albican, Saccharomyces cerevisiae, Neurospora crassa, Fusarium oxysporum, Cochliobolus sati-vu were all inhibited by extracellular polysaccharide, and increases with increasing concentration. Bacteria inhibitory effect were not affected by p H value and have a strong acid tolerance range. While fungi was significantly influenced by p H value,4 Research on extracellular polysaccharide improve memory, anti-fatigue, anti-anoxia test of Favolus squamosusThe results showed that extracellular polysaccharide have significantly affected to memory, anti-fatigue, anti-anoxia of mice.1.169g/kg·d polysaccharide dose group of mice ran the maze time were significantly shorten; 0.500g/kg·d、0.835g/kg·d、1.169g/kg·d polysaccharide dose group of mice loading swimming time were significantly extended; 0.835g/kg·d、1.169g/kg·d polysaccharide dose group of mice anti-hypoxia activity were significant difference / extremely significant; Each dose group of exopolysaccharides feeding mice liver index was not significant influenced, so polysaccharide on mouse liver was non-irritating, non-toxic side effects, develop this new mushroom has a positive meaning.
