At present, although the oligosaccharide production has been a large-scale industrialization development, but most oligosaccharides are produced using enzymatic synthesis or chemical synthesis method to get, there are very few enterprises have extracted directly from plants to obtain it, in the final analysis, the most fundamental reason lies in the higher cost of oligosaccharides extracted directly from plants; in addition,the soybean oligosaccharides are the only widely used as the natural plant oligosaccharides, and the study of red dates oligosaccharides are still under exploration, and there is few report.The advantaged industry in the north of Shaanxi Province---- red dates industry occurred the rapid development over the past 10 years, and has been ranked fifth in the country. But in recent years because of the destruction of the ecological environment, climate and environment have also been greatly affected, the rainfall or drought has increased in the north of Shaanxi Province, resulting in the red dates industry occurred the phenomenon of defective fruit increased, annual output declined, causing red dates cultivation farmers suffered a huge economic losses, so finding red dates industry a new growth point to make up for the farmers for the reduced production t and the increased defective fruit that caused economic losses, or we are able to make the defective fruit from waste to treasure, reprocessing and utilization to increase farmers income will become an important research topic.The value of the significance of this study lies in the optimization of the extraction technology of jujube red dates in the north of Shaanxi Province in oligosaccharide and the research on the effect of its function: lay the foundation for the extraction of red dates natural oligosaccharides; to provide theoretical basis for how to reduce the cost of preparing red dates natural oligosaccharides; to provide new paths and provide a theoretical basis for the new profit growth point of red dates industry in the north of Shaanxi Province.The main contents of this thesis are as follows:1、With petroleum ether as extraction solution for reflux extraction, to study on the optimum about removal of lipids in the red dates. Through a comprehensive analysis of the experimental results, It finally come to this conclusion that the optimum conditions of removal of lipids in the red dates are: the ratio of solid to liquid was 1:10(g/ml) under the temperature of 70 degrees celsius for 2 hours of continuous reflux extraction.2、Using distilled water as extraction solution for reflux extraction, to study on the best process of soluble sugar leaching from red dates. Through single factor and orthogonal experiments results analysis, It finally come to this conclusion that the ratio of solid to liquid was 1:20(g/ml) under the temperature of 70 degrees celsius for 2 hours of continuous reflux extraction, the soluble sugar yield can reach about 75%.3、Using distilled water as extraction solution to extract the soluble sugarof red dates, and then use the Savage method to remove protein, use 95% ethanol precipitation method to remove polysaccharide, and finally by polyacrylamide gel column and activated carbon and diatomite column two different column chromatography make oligosaccharides to be separated from red dates in order to compare, to study on the best technology of oligosaccharides extraction separation of jujube In Northern Shaanxi. It finally come to this determination that ratio of solid to liquid was 1:20(g/ml) under the temperature of 70 degrees celsius for 2 hours of continuous reflux extraction with distilled water, then use two kinds of column chromatography to conduct the separation of oligosaccharides of red dates, the results show that the oligosaccharides yield that by polyacrylamide gel column separation from red dates have a higher ratio than by activated carbon and diatomite column separation from red dates, the highest available oligosaccharides from red dates can reach 572mg/L. 4、In the experiment by measuring the scavenging effect of oligosaccharides in red dates to several different free radical, to study on the antioxidant activity of red dates oligosaccharides.The final experimental results show that:the scavenging rate about red dates oligosaccharides are as follows, the scavenging rate of DPPH was 61.70%; the scavenging rate of hydroxyl was 59.53%, the scavenging rate of nitroso was 66.91%.5、By turbidimetry to mesure two differents liquid bacteria broth turbidity to reflect the culture reproduction of liquid fungus, the differentce between the two medium is whether contains the red dates oligosaccharides, to study on whether the red dates oligosaccharides are beneficial for promot the reproduction of the beneficial bacteria in human and animal bodies. The final experimental results show that the red dates oligosaccharide has the obvious promotion function for the reproduction of the beneficial bacteria in human and animal bodies.
