Visualization Of Binding And Metabolism Of Growh-promoting Peptide By Imaging And Its Affects On Growth-promoting

Growth-promoting peptide is a kind of peptide that may imitate GH activity.Using GH monoclonal antibody as a targets,we have obtained a peptide by immunology and computational biology related technologies.It has been proved that this peptide possess the biological activity and function similar to that of GH. However,its effectiveness remains under in vivo.To visualize the dynamic binding and metabolic characteristics of growth-promoting peptide in tissue and organs.We visualized the distribution and fluorescence intensity of rats in vivo by injected FITC-growth-promoting peptide and FITC-pGH respectively.We found that this peptide and pGH can specifically combined with the main target organs of GH in rats,and showing similar staining whole.But growth-promoting peptide binding degree was weaker than pGH and its the metabolism speed was faster than pGH.In order to verify growth-promoting effect of growth-promoting peptide on animal models.We use experimental rat and one results showed that the rats injected growth-promoting peptide can obviously regulate body weight and length,increasing concentration of OCN(P<0.05),and ALP(P>0.05) in blood.Blood indicator showed that growth promoting peptide regulate body length which is closely related with bone growth.Growth promoting peptide can also regulate body weight which is closely related with the dose.High,middle and low dosage of growth promoting peptide had different impacts on the growth of body weight with different test time. It increased indicated TP(P>0.05) and decreased BUN(P<0.05) in blood, However, IGF-1 level in blood showed not change.Those findings showed that growth promoting peptide regulate body weight gain related closely to promote protein synthesis.Finally,we carried out the experiments to determine activity of growth-promoting peptide on growth-promoting effect in weaned piglets.The experiment group was divided into high dose group and low dose group.The test results showed that:growth promoting peptide can make the ADG of pigs increased,but the variation is not obvious.Results also showed that high dose group was not significant in the early and late(P>0.05),while low dose was significant(P<0.05).It increased blood levels of IGF-1(P>0.05),TP(P>0.05),OCN(P<0.05),the decrease of ALP(P<0.05).The results of this study suggest that: growth-promoting peptide has a promoting effect on piglets that promoting the synthesis of protein and bone growth, but the mechanism still need in-depth study.
