Objective:This study was designed to investigate the influence of the growth, the quality of meat, lipid metabolism and the relative expression of related gene on Schizothorax prenanti Tchang (SPT) adding Citric acid-modified resistent starch (CARS) and Partially Hydrolyzed Oxidized Konjac glucomannan (PHOKGM) in feeding, and evaluate they function, to provide a theoretical basis for the application of CARS and PHOKGM in SPT in the process of breeding.Methods:To add 4.6% starch conducted as the control group (CK), respectively to add 3.2%,1.6% CARS and 3.2%,1.6%,0.8%PHOKGM in the control group replaced the same amount of 10% of the starch as the test group, every group names H-CARS, L-CARS, H-PHOKGM, M-PHOKGM, L-PHOKGM. After feeding 60d, the fish were anatomical, GB was referenced to determine the composition of muscle, using automatic amino acid analyzer to determine the amino acids in muscle. The kits were producted by Nanjing Jiancheng were used to evaluate the content of GLU, TC, TG, HDL-C, LDL-C and so on in the serum through the UV-visible Spectrophotometer. The real-time PCR was employed to detect the relative expression level of the lipid metabolism related genes in the liver, muscle and gut tissue of SPT(for example PPARs, PGC-1α, H-FABP, GPDH. Grelin).Results:1. CARS and PHOKGM didn’t produce significant effects (p>0.05) for the liver index, body fat, abdominal fat index of SPT, can increase SPT weight gain, specific growth, rate of protein efficiency and feed efficiency to varying degrees, while the effects of L-PHOKGM was better than other four groups(p<0.05).2. The two kinds of polysaccharides were no significant effects (p>0.05) for the composed of abdominal and moisture, ash content of back muscles of SPT. However, CARS could be slightly lower the fat deposition of back muscles on SPT (p> 0.05),while PHOKGM could increase fat deposition of back muscle to the varying degrees, and L-PHOKGM increased significantly (p<0.05). L-PHOKGM could slightly increase back muscle protein content (p>0.05).3. The composed of the amino acids in the back muscles and abdominal muscles of SPT were not affected by the two polysaccharides, and there was no difference the composed and content of the amino acids between back muscles and abdominal muscles,the essential amino acids content was about 42% and the content of flavor amino acid was about 39%.Through the chemically score and score, methionine was the first limiting amino acid in muscle of SPT.4. PHOKGM and L-CARS could increase the content of HDL-C (p<0.05). Adding the high dose of the two polysaccharides could reduce the content of TG, TC in serum (p<0.05). Meanwhile H-PHOKGM could reduce the content of LDL-C in serum significantly (p<0.05). Therefore H-PHOKGM improve blood lipid was more prominent than others.5. There was no signidicant influence about CARS affecting the relative expression of PPARa, PPARβ, PPARy in muscle and liver except the expression of PPAR(3 in muscle.The H-PHOKGM and L-PHOKGM increased the relative expression of PPARa, PPARβ, PPARy more significantly than the M-PHOKGM in liver tissue and muscle (p<0.05).PHOKGM significantly reduced the relative expression of PGC-1α in the liver and muscle tissue, while there was no significant effect about CARS. H-PHOKGM and L-PHOKGM promoted the relative expression of HFABP and GPDH in the liver and muscle tissue was more significantly Compared with M-PHOKGM(p<0.05), CARS reduced the relative expression to varying degree.PHOKGM significantly increased relative expression of Grelin in the intestinal tissue(p<0.05), CARS impact on its relative expression not significantly(p>0.05).The two kinds of polysaccharide on the relative expression of GPDH in the intestinal tissue produced unregularity influence.Conclusion:Adding 0.8% PHOKGM in the diet of SPT can promote the growth of SPT so much better than other groups. While adding 3.2% PHOKGM can improve blood lipid more better than others. CARS and PHOKGM have no significant effect on the nutrient component of the SPT meat except adding 0.8% PHOKGM promoting fat deposition of back muscle. Compared with adding 1.6% PHOKGM, the adding of 3.2% and 0.8% can significantly raise the expression of PPARs, H-FABP, GPDH in the liver and muscle, the expression of Ghrelin in intestinal tissue,meanwhile lower the expression of PGC-1α.However the CARS reduces the expression of PPARβ、H-FABP and GPDH and has no significant effect on the other genes.
