Gosling plague, an acute infectious disease is caused by goose parvovirus,especially the 30-day-old goslings and Muscovy ducklings, causing acute onset, high mortality, fast transmission. Morbidity and mortality can decrease with the increasing of age, mainly against within the 7-day-old goslings and Muscovy ducklings. It is divided into three types according to the incidence, most acute, acute and subacute type(Yanzhen and Liu Jinghua, 1997). The main symptoms of the disease are intestinal mucosal shedding, necrosis, intestinal thrombus,dehydration and so on. In 1966, the Hungarian scholar Derszy isolated the virus from goose embryos, and he was the first one who isolated the virus from goose embryos. In 1967 he described it as the flu by mistake, until 1971, when Schettler determined that the disease was caused by another virus. In 1978 it was proposed to be a goose parvovirus. The 1974 World Bird Society named it Derzsy Derzsy(D Derzsy, 1975). In 1995 Zadori first reported the GPV gene sequence, the Muscovy Duck Parvovirus gene sequence and the homology comparison between the two viruses(Zadori Z, 1995).The virus has a certain periodicity. In the pandemic year, the remaining geese won the active immunization, then the following goslings can aslo have natural passive immunity from their parents, so the disease couLd dot broke out in the continuous area. Thus, the morbidity and mortality of the disease was related to the age and the level of specific antibodies of parental goose.Vaccination is an important measure to control the gosling plague. The established methods include PCR, ELISA, indirect immunofluorescence technique, micro neutralization test, but these several detection methods are required with special equipment and time-consuming, not suitable for extension. Therefore, it is urgent to establish a more rapidand sensitive method for virus detection. This study established the polymerase chain reaction,which has the advantages of simple operation, rapid and suitable for the extension.In recent years, gosling plague broke out in some provinces in China and brought huge economic losses to the owners, so the virus get the wide attention of scholars.In the study,we survey local goose breeding status and gosling plague incidence, and the epidemic situation,then do a summary, and make relevant preventive measures in order to raising goose gosling plague disease.In this paper, according to the published GenBank gosling plague VP3 gene sequence information, we design a pair of specific primers. By optimizing the various conditions and eventually amplified, we expected the DNA fragments of 314 bp of the same size. This pair of primer can only amplification of a specific fragment of gosling plague and can be used for detection of gosling plague.According to the investigation of the age or the other basic information, especially immune prevention program,we find the gosling plague occour especially between December and February.The resuLts of survey show that appropriate immunization program is a more effective method to prevent the disease and reduce the probability of the incidence of gosling plague.
