It seems as if the roosters contribute more than 50% of the economic benefits in the chicken breeding farm through their semen. So it is very important to detect the semen quality and use the technique of semen diluting. Now only the semen appearance is inspected, the density and activity, especially the activity, almost was not detected. The semen dilution techniques, which can greatly improve economic benefits, also has not been popularized and applied.For standarding the breeder rooster selection and the semen appearance detection were summarized. Paper study the video microscope system, which is simple, cheap and temperature controlled, was set up and used to detect the semen density and activity, together with the training and detection method. The osmotic pressure, pH and ingredient of YHFB chicken semen dilution, which was studied several years in author’s lab, was optimized and used in 4 large chicken breeder farms. The results showed that:1) The new portable video temperature microscope, which is small and cheap, is very suitable for farm use. The difficulty of chicken semen microscope detection, such as training and result subjective, was reduced with the help of new microscope system, standard density pictures and the objective dynamic activity video. So this equipment promoted the popularization and application of chicken semen microscopy detection technology.2) The methods of detection chicken sperm viability and density, which was written in the current local standard, were modified. The proposed methods were that the vitality and density could be detected simultaneously and there were two choices. The first one was original semen detection, for example, taking semen from 5 roosters in separate tube one by one and detecting immediately. The second one, which could detect lots of samples one time, was collecting semen from different rooster using separate tube and put into box at about 8℃ immediately, and semen sample was diluted with YHFB extender when collected about enough samples (for example 30),then the diluted sample was detected. The activity level must be much lower if the original semen sample was used. The detection temperature for the semen was modified to 32℃ from 37℃ also.3) The chicken semen dilution procedure in the local standard, which was described that the semen and extender must be warmed at the 33℃ water bath, was simplified to only pre-warm the extender according to the specific room temperature. So, it was much more efficiency for the semen dilution technique.4) The highest activity of sperm stored at 5-10℃ was 261mOsm among 340 mOsm.294 mOsm and 261 mOsm. The highest sperm activity stored for 6h at 5-10℃ was the group of the pH 7.38. There was no significant difference in sperm motility if the semen was put first or semen was put first.5) The sperm motility of lycopene group 0.1 mg/mL and 0.2 mg/mL were significantly higher than the control group and lycopene group 0.05 mg/mL. The sperm motility of 0.1 mg/mL lycopene groupwas the best when stored at 5-10℃. The was no significant effect for adding lycopene into the semen dilution when stored at 41℃.6) Using the above technique, through] months of 9 batch incubation experiments, more than 6000 hens (more than 170 thousand pieces of eggs) experiment, the control group, semen and dilution ratio 1:1,1:2 groups, the mean fertilization rate at the 8d hatching was 93.56%.93.36%.93.03%. the variance was 0.10.0.33.0.86 respectively. The egg healthy chick rate were 87.87%.87.38%. 86.58%. the variance was 0.76,0.84,1.34 respectively. There were no significant differences in these three groups.In conclusion, the sperm motility was taked for laboratory testing index, a rooster semen quality detection technology and method of training were integrated innovation.Technology of semen diluent formula was researched and developmented. Through large farm production practice, the farm economic benefit and management level could be greatly improved by the technical system of the paper.
