Feijoa sellowiana is a evergreen shrub or small tree species, belonging to Myrtaceae family. It is native to South Brazil with a secondary dispersion in Uruguay. Numerous studies show that:The feijoa leaves, seeds, peel, pulp and other tissues rich in bioactive plant substances. So it has the effect of antibacterial, antiviral, anti-tumor, anti-cardiovascular disease, liver and kidney, skin care and cosmetics and so on. It is a great resources for the development of medicinal health care.This experiments selected four varieties of feijoa leaves ’Triumph’,’Unique’,’Mammoth’, ’Coolidge’ and its four developmental stages (respectively the size of mature leaves’ 20%, 40%,90% and 100%). Bioactive substances content and antioxidant capacity of leaves were analyzed; and explore the optimum conditions of separation and purification of proanthocyanidins of feijoa leaves. The results were as follows:1.Developmental stage showed a significant impact on the bioactive substances content and antioxidant capacity of feijoa leaves. While, there were no significant difference in the genotype in its impact. In the course of the development of the four varieties of feijoa leaves, Developmental stages of 20% showed the highest antioxidant capacity and the content of total phenols developmental stages, flavonoids and proanthocyanidins.However,with the development of the feijoa leaves, the bioactive substances content and antioxidant capacity of feijoa leaves was gradually decreased. While, ellagic acid in developmental stages of 20% and 90% show the highest content.2. Correlation analysis showed that the content of total phenolic, total flavonoid and procyanidins of feijoa leaves was a significant positive correlation (P<0.01) with the antioxidant capacity. While,the correlation between ascorbic acid and antioxidant capacity was significantly negatively correlated (P<0.05). It showed that total phenols, flavonoids and proanthocyanidins play an important role in terms of feijoa leaves anti-oxidative capacity.3. Determination of procyanidin content by comparing the results of photometric and spectroscopic HPLC results showed:HPLC method for the determination of proanthocyanidins far below the UV spectrophotometry measurement results.The reason presumed that butanol hydrochloride was selectivity in the determination method of procyanidin,such as procyanidin dimer, trimer, multimer etc participate in the reaction. However the HPLC method for the determination of procyanidin dimer having a specific resistance. It can be excluded interference type of polymerization proanthocyanidins. Therefore, the measurement results was small.4. Adsorption and desorption experiments were carried out to screen the suitable macroporous absorbent resin for feijoa leaves proanthocyanidins. State absorption and desorption test of proanthocyanidin are done on the five macroporous resin(AB-8, X-5, D101, S-8, D3520) to study the absorbaility of resin. AB-8 resin is selected as optimal packing.The test on AB-8 indicates that the resin saturate on the condition of pH was 4 and elution solvent is 60% ethanol. The sample flow rate is 2ml/min, the maximum amount of the sample with the mass ratio of macroporous is 20:1 (v/m), the ratio of diameter to height is 1:6, the eluent flow rate was 1ml/mm, eluent amount 4.3BV (BV means column volume). Proanthocyandins detection products yield 87.3%, purity 44%.
