Effects Of Complex Chinese Crude Drug "Zhukangsan" On The Cellular Immunity Of Weaned Piglets Artificially Challenged By PRRSV

This study investigated the different dosage of complex Chinese crude drug "Zhukangsan" on the levels of immune globulin and cellular immune function of weaned piglets artificially challenged by PRRSV, aimed to reveal the prophylactic effect of complex Chinese crude drug "Zhukangsan" on PRRSV, while explored the best added dose and its mechanism of action, providing a scientific and theoretical basis for application in the production of the livestock.Forty piglets of 28-day-old, no clinical symptoms, weight approximation were divided randomly into 4 groups, each group of 10 head. After one week adaptation to dietary daidzein, Group III and IV were given drug (Zhukangsan) at 0.5% and 1.0% in the forage at 36 to 39-day-old, respectively, then all groups except group I were inoculated PRRSV SCM at 40-day-old. In 40,45,50,55 and 60 days of age in each group were randomly selected 4 piglets of vena cava blood testing piglets lymphocyte subsets in peripheral blood, lymphocyte transformation rate and red cellular immune function and choose 4 piglets slaughter at 50 and 60 days of age randomly, detecting of splenic lymphocytes cycle and apoptosis.Results:(1) "Zhukangsan" has reduced the quantity of G0/G1 lymphocytes, increased the quantity of S and G2+M lymphocyte and lymphocyte PI value function (P< 0.01 or 0.05).0.5% Chinese medicine group phase G0/G1 lymphocyte number is better than 1%(P< 0.01), better than the 0.5% group values of 1% traditional Chinese medicine group stage S,G2+M lymphocyte number and PI (P< 0.01). The test results show that the compound Chinese medicine "Zhukangsan" has the effect of promoting proliferation of splenic lymphocyte, recommend the use of 0.5% dosage.(2) "Zhukangsan" can reduce the apoptosis of splenic lymphocytes rate effect, can effectively inhibit the increased splenic lymphocytes apoptosis rate, and the effect can be maintained for a long time, particular with 1% additives beam (P< 0.01 or 0.05).(3)"Zhukangsan" can improve T lymphocyte subsets and CD3+, CD3+CD4+, CD3+CD8+ and CD4+/CD8+, the percentage of the role, comprehensive analysis, "Zhukangsan" 1% add a dose effect is better than that of 0.5% dosage (P< 0.01 or 0.05).(4) "Zhukangsan" can effectively improve piglet peripheral blood lymphocyte transformation rate, and maintained for a long time, among which 1% added dose effect is the best (P< 0.01).Conclusion:Compound Chinese medicine "Zhukangsan" can reduce infection PRRSV piglet the number of spleen lymphocytes of G0/G1 phase, S phase and increase the quantity of G2+M lymphocyte and PI lymphocyte percentage value, reduce the apoptosis of spleen lymphocytes, enhance the lymphocyte subsets in peripheral blood CD3+, CD3+CD4+, CD3+CD8+, CD4+/CD8+ and the percentage of lymphocyte transformation rate, improve the peripheral blood erythrocyte complement receptor rosette rate and effect to reduce red blood cell immune complex rosette percentage of weaning piglets, thus promoting immune organ development of the spleen, promoting lymphocyte proliferation, reduce the apoptosis of lymphocyte and improve the cellular immune function of the body. Obviously the cellular immune function in piglets added 1% feed is better than add 0.5% feed "Zhukangsan".
