Ancient Platycladus orientalis and Sophora japonica which accumulated thousands of years of vicissitudes, are unique historical and cultural products and ecological heritage of China. In order to provide practise theories and technologies for well protection those kinds of ancient trees,, this study collected the ancient Platycladus orientalis and Sophora japonica samples from Yangling, Zhougongmiao and Huangdiling area, respectively, to isolate flora endophytic fungi, morphological identification and molecular systematics techonlogies were employed to classify endophytic fungi’ taxon placement. Endophytic community structure and features are furthure analysized by statistic and PCA(principal component analysis)methods, so as to clearify the difference between those two ancient trees’ endophytic fungi coummunities in aspects of composition, structure characteristics and the influence causing by the age factor. Achievements are made as follows:1. 705 strains of endophytic fungi which were got from two trees belong to 26 genera, 36 species, respectively. Strains of 699 are ascomycetes belonging to 24 genera, accounting for 99.14% of all strains. And 6 strains are basidiomycetes belonging to 2 genera, accounting for 0.86% of all strains.2.The community composition between endophytic fungi in ancient Platycladus orientalis and Sophora japonica both have large differences with the similarity coefficient less than 0.5. The endophytic fungi diversity coefficient of ancient Platycladus orientalis in Zhougongmiao(H’=2.11) is higher than that in Yangling(H’=1.75), and diversity coefficient of ancient Sophora japonica in Zhougongmiao(H’=1.95) is higher than that in Huangdiling(H’=1.61), which displayed the diversity of endophytic community was influenced by these 3 different sampled sites.3.PCA analysis shows that, the age of a tree is also one of the major factors of the composition and distribution. The greater the age of the same host tree is, the higher the endophyte fungi diversity index and richness index is, and the higher the complexity of the composition is.4.These two kinds of ancient trees still share two kinds of floras, Alternaria and Fusarium are their common endophytic isolates. The unique floras in blades of the ancient Sophora japonica are Colletotrichum(RF=14.09%), Phomopsis(RF=2.71%), Thielavia(RF=1.08%) and Sordaria(RF=1.21%), whereas, the ancient Platycladus orientalis breed Xylaria(RF=5.95%), Acremonium(RF=1.49%), Cladosporium(RF=1.19%), Pestalotiopsis(RF=3.87%), Coprinellus(RF=0.63%),and so on isolates, and the composition of the endophyte fungi community may be the important factors associated with the health of those two ancient trees.
