In this experiment, F4 strain of Flammulina velutipes was used as testing material. Studying on the degradation of their main components in different periods and the dynamic changes of activities of some relevant enzymes, investigating the impacts of exogenous cellulase on Flammulina velutipes’mycelial growth and biological efficiency and discussing the feasibility of applying exogenous cellulase to Enoki mushroom by adding complex exogenous cellulase of different concentrations to the substrate. And this offers a reference for Enoki mushroom’s high-efficient cultivation. The main results as below:1. Exogenous cellulase has more or less inhibitory effect on the rate of mycelium growth. Growth rate of exogenous cellulase of low concentrations were slightly lower than that of the control group, while higher concentrations of it inhibit mycelium growth rate significantly; Exogenous cellulose has significant effects on improving mycelium biomass, and the mycelium biomass of treatment group A1 and A2 of respectively 54.8% and 32.0% higher than the control group.2. Added exogenous cellulase to the liquid fermentation medium of Flammulina velutipes got the pH of the fermented liquid decreased obviously, but the sugar content, soluble protein content of it have increased significantly. The contents of sugar and soluble protein in treatment group A3’were the highest which were 0.79 mg/ml and 3.24 mg/ml higher than the control respectively. At the moment, exogenous cellulase has promoting effect on 5 types of extracellular enzyme activity, including CMCase, HCase, Amylase, Pectinase, and Laccase.3. Added exogenous cellulase to the substrate of Flammulina velutipes had a great influence on extracellular CMCase activity, but little effects on other 5 kinds of extracellular enzyme. At the beginning of mycelium growth, the activity of exstracellular cellulase in the treatments which add exogenous cellulose after sterilization are increasing with the enzyme concentration increases, the extracellular CMCase activity of CK1, CK2 in the treatments adding exogenous cellulase before sterilization changes like a double peak curve. The biggest peak activities of Extracellular HCase, FPase Pectinase were at the harvest time, while Amylase and Laccase at the time of mycelium growth have higher enzyme activities.4. In different growth periods, the content of reducing sugar were different, and that of the groups which adding exogenous cellulase before the sterilization contain more reducing sugar than the control group; different concentrations of composite cellulose matrix content of soluble proteins of the overall trend, fruiting body maturation levels reach the maximum, highest content in A3,up to 48.08 mg/g, decreased gradually after harvesting.5. Addded exogenous cellulase has an effect on promoting the lignocellulose decomposited of the substrate. Processing B1 has the highest degree in the degradation of wood fiber, reached 27.01%,1.91% higher than CK2. Cellulose and hemicellulose degradate mainly in the fruit body growth stage, and the degradation of lignin is mainly in mycelial growth stage.6.The cultivation results of adding exogenous cellulase to the substrate of Flammulina velutips showed that exogenous cellulase has inhabit effect on the mycelium growth rate, and lower concentrations of exogenous cellulose has a certain role in promoting Enoki mushroom production. Treatment B1 has significant advantages over fruiting body of agronomic traits, yield and nutrient composition.The biological efficiency of B1 reached 123.82%,26.93% higher than the control,which will be good for high-efficient cultivation of Enoki mushroom.
