Chinese orchid refers to part of the terrestrial orchids of Orchidanceae Cymbidum originated in China and it mainly includes Cymbidium goeringii, Cymbidium faberi, Cymbidium ensifolium, Cymbidium sinense and Cymbidium kanran.The reasearch took Cymbidium goeringii and Cymbidium faberi as the test materials, which was mainly consisted of content at two aspects:firstly, achieving the purpose that Cymbidium goeringii blossoms during Chinese Lunar New Year through the treatment of temperature and exogenous GA3; Secondly, studying each procedure of Cymbidium faberi of tissue culture from germination to adult seedling and focused on studying the influence of vaccination site, light intensity and the method of culture factors on rhizome multiplication,optimized tissue culture and rapid propagation system for Cymbidium faberi and made the preliminary exploration on one-step seedling formation of Cymbidium faberi. Cost and feasibility were taken into account in test procedures to provided reference for large-scale production practice.The main research results were as follows:1. After proceeding natural low-temperature treatment for 30 days and placing it in glass greenhouse for 30 days with the average temperature 17~20℃ and air humidity 75%~85%,the flowing rate of Cymbidium goeringii during Chinese Lunar New Year reached 78.57% and its scape height reached 12.64 cm, which was the optimum treatment combination to promote florescence in advance and improve ornamental value.2. The treatments of spraying GA3 and smearing scape could not play an effective role in regulating the flowering time of Cymbidium goeringii, especially high-density single spraying always causing leaf damage and lowering overall appreciation. But we found that proceeding natural low-temperature treatment for 30 days and then spraying the Cymbidium goeringii with 90 mg/L agricultural GA3, the flowing rate reached 88.57% and its scape height reached 13.90 cm.3. The germination abilities of different seeds of Cymbidium faberi was different significantly,the optimum mediums was different as well. In this research,the seeds of chengmei’x’biancao’showed the best germination abilities and the optimum medium for seed germination was hyponexl 5g/L+NAA 5mg/L+sucrose 20 g/L+agar 7.5 g/L+activated carbon 2 g/L+peptone 3 g/L.4. The medium hyponexl 5g/L+NAA 5mg/L+sucrose 20g/L+agar 7.5g/L+a ctivated carbon 2g/L+peptone 3g/L was suitable for growth and multiplication of the rhizome; the head segment rhizome had the strongest multiplication capa city while the middle and end segment did not have significant difference; The illumination 6000 Lx was most conductive to rhizome multiplication and rhizo me was undergrowth under dark condition; solid static culture method was mos t beneficial to rhizome multiplicatio,no significant difference was found in stat ic liquid culture and shaking culture.5.1/2MS+NAA 1mg/L+BA 1 mg/L+sucrose 20g/L+agar 7.5g/L+activated ca rbon 2g/L was a relative appropriate medium for one-step seedling formation,w hich realized rhizome multiplication and seedling induction of Cymbidium faber i at the same time.6. SOD, POD and CAT cooperated with each other to adjust the differenti ation and development process of the rhizome.The results showed that low leve ls of SOD and POD activities and high level of CAT were associated with rhi zome multiplication, while the bud rate of the treatment of high-activity SOD and low-activity POD was higher.
