Sophora alopecuroides(SA) belong to the pure natural Chinese herbal medicine and its whole plant taste bitter, cold and toxic. Its bean plant as a part of SA which used as a folk medicines to treat the heat clearing,flow field and the parasite.Modern medical and veterinary research showed that SA has anti-microbial, anti-inflammatory, anti-cancer, insecticidal, enhanced immunity and other pharmacological effect.This experiment mainly studied Sophora alopecuroides extracts(SE) had antibacterial, anti-inflammatory, immune function and its effect on the toxicity research in mice. 1. Isolation of pathogenic bacteria from dairy cow mastitisThis study was to survey the types of pathogenic bacteria and isolated pathogenic bacteria from dairy cow mastitis.Take 46 milks sample with sterile from Shaanxi Baoji in dairy mastitis cows to isolate and identify pathogenic bacteria. We identified 4 pathogenic bacteria, such as Streptococcus dysgalactiae, Staphylococcus aureus, Staphylococcus epidermis and Escherichia coli. 2. SE inhibited pathogenic bacteria in vitroThe purpose of this study was to study on the anti-bacterial effect of SE on mastitis pathogens in vitro. Methods the anti-bacterial effects of SE were inhibited by Oxford cup and tube double dilution method in vitro. The minimum inhibition concentration(MIC) of SE to Staphylococcus aureus, Staphylococcus epidermis, Streptococcus dysgalactiae and Escherichia coli was 0.585、2.340、2.340、4.687 mg/L,while the minimal bactericidal concentration(MBC) was 9.375 mg/ L, respectively. This study indicated that SE had obvious bacteriostatic effect in vitro. 3. SE has anti-inflammatory effectThe present study was designed to evaluate the effects of SE on anti-inflammatory in mice. The anti-acute inflammation was evaluated by xylene induced ear edema and using egg white-induced paw edema in mice.The anti-chronic inflammatory activity was evaluated by cotton ball granular in mice. SE high, medium and low dose group were significantly decreased the mice auricle swelling degree, the auricle swelling rate were 82.61%, 65.22% and 42.83%, respectively; SE has markedly inhibit inflammation(toes swelling), which caused by egg white;SE high, medium and low dose group could inhibit granulation tissue induced by cotton ball in mice and proliferation inhibition rate was 54.18%, 35.45% and 39.76%, respectively.SE has visible anti-inflammatory effects. 4. SE could improve immune functionThis study was designed to evaluate the effects of SE on immune function in mice. The effects of SE influenced immune organs weight. The effects of SE on cell immune function were immunized by sheep red blood cells(SRBC). The specific anti-body(Ab) titer was assessed by SRBC test. The humoral immune function was assessed by determining half value of hemolysis(HC50) in mice. The increase weights of the spleen and thymus indexes suggested that SE had significant effect on the major immune organs in mice. SEhigh, medium and low dose groups had significant effect on mice cellular immune function and its immune response(foot plantar thickening) were 82.38%, 59.07% and 59.33%, respectively. SE high, medium and low dose groups had obviously improved the SRBC Ab titer, which antibody titer respectively were 1:1280、1:1280、1:640. SE high, medium and low dose groups can obviously improve half serum hemolysis value(HC50) in mice and HC50 values were145.10±4.26, 136.12±6.15, 133.70±6.09. SE could enhance immune functions in mice. 5. The effects of SE on acute toxicityThis study was designed to study the effects of SE on acute toxicity in SPF mice. The mice were garaged with SE. The toxic and adverse reactions were observed and recorded. The improved karber method was used to calculate the median lethal dose(LD50) values and the 95% confidence limits. The results showed that the LD50 value of SE was 144.63 mg/L,the 95% confidence limits were from 109.88 to 191.89 mg/L, and SE belongs to the medium toxic drugs.
