Study For The Effect Of Mitigating Heat-stressed In Cows With Se-enriched Ligustrum Lucidum Fruit

Heat stress is a GAS caused mainly by high temperature and high humidity. According to the basic theory of the etiology and pathology of Chinese traditional veterinary medicine and diagnosis of dialectical, heat stress belongs to yang deficiency syndromes because of the evil high fever, and Ligustrum lucidum possesses the function of nourishing the kidney. Our previous study showed that Se-enriched ligustrum lucidum fruit could effectively reduce the mortality of mice suffering from acute heat stress mice, and relieve the extent of liver injury caused by heat stress in mice. It not only can Improve the immunity of goats but also ease milk production decline of heat stress goat. We also made research on the effects of three kinds of effective components of Se-enriched ligustrum lucidum fruit on goat’s peripheral blood lymphocytes proliferation activity and cytokines secretion. Therefore we performed a research on the effect for anti-heat-stressed research using Se-enriched ligustrum lucidum fruit in dariy cows. To observe the effects of Se-enriched ligustrum lucidum fruit on production performance of cows and Hematology and serum biochemical indexes. We also extract protein from Se-enriched ligustrum lucidum fruit to study its effect on peripheral blood lymphocyte,proliferation and cytokines secretion in cows。Test 1 The effects of Se-enriched ligustrum lucidum fruit on dairy cowsUsing Ligustrum Lucidum Fruit rich in Se to control heat shock. Chose 50 dairy cattles which are similar in ages, parity and milk yield and put them into 5 groups named as HS, MS, LS, P and C group. The dosage of group HS, MS, LS is 60g,40g and 20g Ligustrum Lucidum Fruit rich in Se per day. The dosage of P group is 40g Ligustrum Lucidum Fruit per day. The C group was subjected to no disposal. The dosage regimen is that the dairy cattles were given medicine at the 15:00 in the first three day of each period of ten days.The milk yield of the three group which was feed Ligustrum Lucidum Fruit rich in Se decreased by 23.91%,28.87% and 25.23%.It’s significantly lower than the other group which is 39.19% and 36.55%(P<0.05). The milk protein content of MS group and P group is obviously higher than usual(P<0.05). The ACTH and Cortisol level of MS group had a significant reduction(P<0.05), the T3 level of MS group went back to normal. The HSP70 level of HS and MS group were significantly improved(P<0.05).The.antioxidation index (SOD and GSH-PX) of MS and P group were significantly enhanced (P<0.05).The IFN-y level of HS/MS/LS group were significantly improved (P<0.05) and that in the group 2 was highly significant in differences (P<0.01). The results indicated that Se enrich Ligustrum Lucidum Fruit has a positive effect on resisting heat shock.Test 2 Effect of Se en-rich Ligustrum Lucidum Fruit Protein on proliferation of PBL and cytokine secretion of cowsExtract protein from Se enriched Ligustrum Lucidum Fruits to research its effects on proliferation of PBL and cytokine secretion and use pre-HPLC to separate SLLPF from the crude extract of Se-enriched ligustrum lucidum fruit. The result shows that two kinds of protein both can promote lymphocyte proliferation under the stimulation of ConA, and it increase with the increase of protein concentration. When the concentration of the protein is higher than 50ug/ml, the changes of SLLPF group and the SLLP group are significant compared with the control group (P<0.05), When the concentration of the protein is higher than 400μg/ml, the effect of normal Ligustrum Lucidum Fruit Protein are significant compared with the control group (P<0.05) and the effect of Se-enriched Ligustrum Lucidum Fruits protein is better than the effect of Ligustrum Lucidum Fruits protein and the difference are significant(P<0.05). The concentration of IL-2,IL-4,IL-6 and IFN-y of Se-enriched group were significant higher than the control group(P<0.05), the IL-4 concentration was very significant higher than the control group(P<0.01). The IL-4 and IFN-y concentration of LLF (Ligustrum Lucidum Fruits protein)group were also significant higher than the control group(P<0.05). It is shows that the Se enriched Ligustrum Lucidum Fruits protein can improve the activitiy of the PBL and the secretion of cytokines. It is indicates that the SLLPF is one of the effective parts of Se en-rich Ligustrum Lucidum FruitConclusion:Adding 20～60 g/case/d dose of Se-enriched ligustrum lucidum fruits to the basal diet can alleviate the decreased milk production caused by heat stress(increase 2.89%),of witch 40 g/case/d Se-enriched ligustrum lucidum fruits works best and its effect is better than common ligustrum lucidum fruits. The function mechanism of Se-enriched ligustrum lucidum fruits is regulating the secretion of hormones which are related to heat stress,such as ACTH, COR et al by the HAP axis. It also can improve immune function by increase the expression level of the cytokines. which make the body maintain good homeostasis and then enhance its immune function. The Se-enriched ligustrum lucidum fruits protein is the main active ingredient of the Se-enriched ligustrum lucidum fruits.
