Study For The Effect Of Mitigating Heat-stressed In Goats With Se-enriched Ligustrum Lucidum Fruit

According to the basic theory of the etiology and pathology of Chinese traditional veterinary medicine and diagnosis of dialect ical, heat stress belongs to yang deficiency syndromes because of the evil high fever, and Ligustrum lucidum possesses the function of nourishing the kidney; our previous study showed that Se-enriched ligustrum lucidum fruit could effectively reduce the mortality of mice suffering from acute heat stress mice, and relieve the extent of liver injury caused by heat stress in mice, not only increases the concentration of total serum TP,GLO and ALB, but also enhance immunity function of mice. Therefore we performed a research on the effect for anti-heat-stressed research using Se-enriched ligustrum lucidum fruit in goats and conducted the safety trials and thermal stress effect for Se-enriched ligustrum lucidum fruit,to explore the effect and mechanism of the anti-heat-stress of the Se-enriched ligustrum lucidum fruit in goats,in which we observed production performance,hematology, serum biochemical parameters and peripheral blood lymphocyte proliferation, cytokine secretion, and antioxidant activity in goats.Test 1 The Security Test of Se-enriched Ligustrum Lucidum Fruits in GoatsThis study was designed to observe the security of Se-enriched ligustrum lucidum fruits in goats.30 adult goats producing milk were randomly divided into five groups, each goat in the group C was fed with the basal diet, each goat in group CLL was fed with the basal diet with common ligustrum lucidum fruits 30g/d, each goat in group LSLL, MSLL and HSLL were respectively fed with the basal diet with lOg/d,30g/d,50g/d of Se-enriched ligustrum lucidum fruits, trial group at the start of the first test 5d administration for 5 days. Detection of goat milk, blood WBC, RBC, HGB content and serum the levels of TP, ALB, ALT, AST, CK, LDH. The results showed that Se-enriched ligustrum lucidum fruits and common ligustrum ligustrum fruits both can increase the milk yield. In test 1-7d,test 22-38 d milk yield growth in descending order:group LSLL>group MSLL>group HSLL>group CLL>group C; group CLL can significantly improve the RBC of goats (P<0.05), group MSLL, group CLL could significantly improve the HGB content of goats (P<0.05); group LSLL, group MSLL, group HSLL and group CLL could significantly improve goat’s serum TP content (P<0.05),group LSLL, group MSLL and group CLL could significantly improve the goat’s serum ALB content (P<0.05), group LSLL can markedly reduce serum AST activity (P<0.05),it does not significantly affact the WBC, ALT, CK, LDH activity of goats to add high selenium content of ligustrum lucidum into daily ration (P>0.05). It is demonstrated the dietary Se-enriched ligustrum lucidum fruits can increase the milk production of goats, meanwhile also improve the blood RBC and HGB content and increase the levels of serum TP, ALB, reducing goat serum AST activity, but it showed no significant effect on WBC, ALT, CK, LDH activity. These results showed that adding≤50 g/case/d dose of Se-enriched ligustrum lucidum fruits to the basal diet in conjunction 5d is safe to goats.Test 2 Effect of Se-enriched Ligustrum Lucidum Fruits in Heat-stressed GoatsIn order to observe the effect of Se-enriched ligustrum lucidum fruits on performance, hematology and serum biochemical indexes in goats in this test.30 goats producing milk were randomly divided into five groups, each goat in the group HS was fed with the basal diet, each goat in group CLL was fed with the basal diet with common ligustrum lucidum FruitslOg/d, each goat in group LSLL, MSLL and HSLL were respectively fed with the basal diet with 5g/d, 10g/d,20g/d of Se-enriched ligustrum lucidum fruits. The test started from July 10 to August 27 in 2014,and trial group mixed feeding with the basal diet administered every ten days to three days ago, the administration for four times. Detection of goat milk, blood WBC, RBC, HGB content and serum the levels of TP, ALB, ALT, AST, LDH,IL-2,IFN-y,IL-6,SOD,GSH-Px,ACTH and COR. The results showed that diets supplemented with Se-enriched ligustrum lucidum fruits can increase heat-stressed goats’milk production, and milk production decline rate ascending order:group MSLL> group LSLL> group HSLL> group CLL>group HS, while group MSLL increased by 2.89% than in the non-heat stress period, and significantly higher than the group HS decreased 10.07%(P<0.01);and Se-enriched ligustrum lucidum fruits significantly increased fat content(P< 0.05),improved the protein content of milk and serum TP,ALB content; HSP70 expression of group HSLL and LDH activity of group LSLL were significantly lower than group HS in 28 d,IL-2,IFN-y content and SOD activity of group LSLL were significantly higher than group HS(P<0.05),COR concentration of group MSLL,HSLL,CLL was significantly lower than group HS in 14 d(P< 0.05),impacting of lactose, total milk solids, non-fat milk solids,WBC,RBC,HGB,ALT,AST,IL-6 was not obvious(P>0.05).The results demonstrated Se-enriched ligustrum lucidum fruits can improve the milk production of heat-stressed goats by acting endocrine-immune regulation network system and improving metabolism, especially adding 5-10 g/case/d dose of Se-enriched ligustrum lucidum fruits to the basal diet can reach the best effect.Test 3 Effect of Se-enriched Ligustrum Lucidum Fruits on Goat Peripheral Blood lymphocyte Proliferation ActivityIn order to investigate the effect of Se-enriched ligustrum lucidum fruit in goats and some indexes, such as peripheral blood lymphocyte proliferation, cytokine secretion, and antioxidant activity were observed. To determine the peripheral blood lymphocyte proliferation activity of Se-enriched ligustrum lucidum fruit polysaccharides, proteins, oleanolic acid in goats by MTT, and cytokines IL-2,IL-4,IL-6,IFN-y level and SOD activity of its culture supernatant were detected. The results showed that Se-enriched ligustrum lucidum fruit polysaccharides, protein, oleanolic acid in vitro had a role in promoting ConA stimulation of peripheral blood lymphocytes proliferation, and the effect of Se-enriched ligustrum lucidum fruit protein, oleanolic acid components were better than common ligustrum lucidum fruits, the corresponding concentrations was significantly different which respected 63.64% and 40.00%.Se-enriched and common ligustrum lucidum fruits were significantly increased activity of SOD in peripheral blood lymphocytes(P< 0.01),and the effect of Se-enriched ligustrum lucidum fruits was significantly better than common. Se-enriched ligustrum lucidum could stimulate lymphocytes to produce IL-2,but with the control group the difference was not significant(P>0.05).The results showed that Se-enriched ligustrum lucidum polysaccharides, protein, oleanolic acid have a role in promoting peripheral blood lymphocytes proliferation,but also significantly improve their antioxidant capacity.Conclusion:Adding≤50 g/case/d dose of Se-enriched ligustrum lucidum fruits to the basal diet in conjunction 5d is better security for goats; and alleviate the decreased milk production caused by heat stress(increase 2.89%),of witch 5-10 g/case/d Se-enriched ligustrum lucidum fruits is better and the effect is better than common ligustrum lucidum fruits. The function mechanism of Se-enriched ligustrum lucidum fruits is reducing the heat stress of elevated ACTH,COR and upped the heat stress decreased IL-2、IL-6、IFN-γ levels by the HAP axis, which make the body maintain good homeostasis and then enhance its immune function;while increasing serum TP,ALB content, GSH-Px, SOD activity and decreasing LDH activity,HSP70 expression level, so that the body reduces the stress compensation, keeps normal metabolism and maintain a more normal period of milk production.
