Melatonin could induce cashmere growth, increase cashmere production, improve cashmere quality in cashmere goat. However, its regulatory mechanisms haven’t been reported. Hair and cashmere can derive from primary and secondary hair follicle, the research took hair follicles of Inner Mongolia cashmere goats as research objects, added melatonin to a culturing model of isolated hair follicles in vitro, observed hair follicles’ growth condition, carried out transcriptome profiling of hair follicles which after cultured using RNA-seq, and excavated genes related to hair follicle growth and development. The results demonstrated that,1. The number of hair follicles which were isolated by collagenase and incision cutting method was more, hair follicles’morphostructure was clear, dermal papillas were well growth, impurities of fat were less, this method was adequate for isolating hair follicles of cashmere goat.2. The promoting effect of melatonin on primary hair follicles was significant(P<0.01), the promoting effect of melatonin on secondary hair follicles was nothing, primary hair follicles’ growth rate reduced with culture time extending.3. According to transcriptome research of hair follicles which cultured in vitro, total52418238,63697465,47806587and50152649reads were generated in PF_K, PF_MT, SF_K, SF_MT, respectively. A total of1408differential genes were identified between PF_MT and PF_K,1471differential genes were identified between SF_MT and SF_K,3477differential genes were identified between SF_K and PFK,3397differential genes were identified between SF_MT and PFMT.4. A quantity of differential genes between PF_MT and PF_K were predominantly enriched in drug metabolic process, structural constituent of ribosome and cytosolic large ribosomal subunit GO terms; A quantity of differential genes between SFMT and SF_K were predominantly enriched in oxidation-reduction process, oxidoreductase activity and extracellular region GO terms; A quantity of differential genes between SF_K and PF_K were predominantly enriched in cell adhesion, structural molecule activity and intermediate filament GO terms; A quantity of differential genes between SF_MT and PF_MT were predominantly enriched in cell adhesion, cell-cell adhesion, structural molecule activity and intermediate filament GO terms. 5. Pathway analysis results presented:Plenty of differential genes between PFMT and PFK were predominantly enriched in Ribosome, plenty of differential genes between SF_MT and SF_K were predominantly enriched in Butanoate metabolism, plenty of differential genes between SF_K and PF_K were predominantly enriched in Malaria, plenty of differential genes between SF_MT and PF_MT were predominantly enriched in Systemic lupus erythematosus and Arachidonic acid metabolism. Wnt, Shh, MAPK, NF-kB, TGF-β, JAK-STAT and Notch signaling pathways involved in hair follicle growth and development were found out in pathways which differential genes were enriched. The functions of many genes in hair follicle growth and development were not reported in pathways yet. They, as candidate genes in melatonin promoting hair follicle growth, need to be further investigated.
