In this study, we separated and cultivated cashmere goat hair follicle stem cells by two methods, one was the two-step enzyme digestion, another was differential adhesion. Growth curves and colony forming efficiency of passaged cashmere goat hair follicle stem cells were determined. The preliminary identification of hair follicle stem cells by using morphology, induced differentiation and gene detection. We discussed the most suitable concentration of melatonin for cashmere goat hair follicle stem cells. And we cloning the melatonin receptors gene cDNA by RT-PCRThe main results are as follows:1, Cashmere goat hair follicle stem.cells were uniform, volume small,paving stone-like, active proliferative capacity, shape of growth curve and can be induced to differentiate into adipocytes, osteoblasts and neural like cells. They could espress the piuripotency genes of Oct4, Sox2, Nanog, Sox9, integrin-β1, CD34, CD200and CK15.2, The result indicated that500pg/ml is the most suitable concentration of melatonin for cashmere goat hair follicle stem cells.3, The MTNR1A, MTNR1R and RORa cDNAs were cloned from Cashmere goat hair follicle stem cells. We acquired a part of sequeces of the MTNR1A, MTNR1R and RORa for518bps,477bps and551bps,Sequence analysis about the melatonin receptors indicates that there were high homologies of nucleotide acid between goat and other bovidae mammalian.
