In this study, petiole and stem, leaves of Peperomia obtusifolia were used as explants, to carry out the study of Peperomia obtusifolia organogenesis and somatic embryogenesis and studied on the domestication and seedling test. The main results are as follows:1. Peperomia obtusifolia organogenesis approach1.1 The results showed that the stem and petiole were the best explants.The best combination of sterilization:Petiole in 0.1% mercuric chloride sterilization 7min; stem section 0.1% mercuric chloride 9min+75% alcohol 30s.1.2 The study found that induction medium of leaves and stems of the best adventitious buds were 6-BA 2 mg/L+IBA 0.5mg/L,induction medium of petiole was 6-BA 2 mg/L+IBA 0.2mg/L. The results showed that the adventitious bud proliferation group the most suitable medium was 6-BA 2mg/L+GA3 0.3mg/L+IBA 0.3mg/L. The most suitable medium for the growth of single bud stem segment was 6-BA 0.5mg/L+GA30.3mg/L.1.3 Study shows that the most suitable rooting medium is 1/2MS+IBA 0.2mg/L+AC lg/L. And found by adding different concentrations of perlite can reduce the gas that rooting effectively.2. Study of Peperomia obtusifolia embryoid2.1 Induction medium for Peperomia obtusifolia,Leaves Callus was 6-BA lmg/L+2.4-D 3mg/L.Under both light and dark culture, the embryos could be formed.2.2 The results showed that the optimum medium for the proliferation of somatic embryos was 6-BA 2mg/L+NAA 0.5mg/L+sucrose 30g/L, and the effect on the proliferation of the somatic embryos was 6-BA>NAA> sucrose. In the culture mode, the liquid is better than the solid.2.3 In the experiment of embryo differentiation into Plantlets, the results showed that 6-BA 1.5mg/L+IBA O.lmg/L was the best differentiation medium. The inoculation rate of mass (2-3) was the highest. Suspension culture can also be differentiated into seedling on its medium.3. Peperomia obtusifolia Seedling Domestication and Seedling Technology ResearchThe results showed that the first closed bottle 5d, after opening 2d, transplanting survival rate is high; in the humidity, it was found that once every three days, the survival rate of transplanted. Perlite:peat =1:3 for the best transplanting survival rate. The seedling test showed that spraying 2000 times more flower fertilizer No.1, seedling index was significantly higher than other groups, supplemented with the best strong sprout mainly by spraying different concentrations of fertilizer available. The results showed that application of 1500 times and 2000 times of Huaduoduo 1 hao(20-20-20) and 800 times of Huaduoduo 8 hao(20-20-20) seedling index was significantly higher than the other groups, the treatment group is best treated satrong sprout.
