The Construction Of Peperomia Obtusifolia Leaf On High Efficiency Propagation System

Peperomia obtusifolia is a kind of Peperomia. This plant owns ornamental and medicinal values, at present, it most propagated by cuttings and plant distribution method, since it’s too difficult to meet the market demand for peperomia obtusifolia with the low propagation coefficient, it has important practical significance to explore the rapid propagation for peperomia obtusifolia.In this study, the leaf of peperomia obtusifolia were chosen as experimental materials, and the key technology in peperomia obtusifolia tissue culture, such as pre-sterilization, hormone, plant growth regulator, the bud induction, rooting. Finally established a regeneration system of peperomia obtusifolia by using the technology of tissue culture for factory production. The main results of this study are as follows:1、The best method for sterilize of explants:it was dip in 75% ethyl alcohol for 20s after water flushed 10 min, then agitating in a solution of 0.1% HgCl2 for 4 min, and finally rinsed four times with sterile distilled water.2、The results of induction of callus and adventitious bud revealed that anxin and cytokinins concentration was the main factors that influence callus induction and diversification. The growth and differentiation state of callus were better in the culture medium with 6-BA1.0mg/L and NAA1.0mg/L. Added the anti-browning agent VC 60mg/L can effectively reduce the rate of explants browning, when inoculating the explants, compared to the oblique, horizontal inserting on culture medium will increase the appearance of the explants browning, but the rate of callus induction and the number of adventitious buds is higher.3、In subculture, the suitable growth hormone combination and density was MS+6-BA2.0mg/L+NAA0. 1mg/L.4、Roots could be easily induced, but the quantity and the quality of the resultant roots on different media was obviously different;5、In subculture and rooting culture of the peperomia obtusifolia, we can substitute white granulated sugar for sucrose, tap water for distilled water.6、The best time in domesticate of rooting seedlings is 3days.
