| Chinese herbal medicine with small toxic,green and no residue characteristics has become a hot research field of feed additives,especially the study of Chinese herbal medicine additive to improve animal body’s disease resistant ability,which gets more and more attentions of people.In order to explore the effect of No.3 Chinese herbal medicine additive on immune index of piglets to improve disease resistance and provide a scientific basis for green and healthy development of the pig industry.The test select 60 15-dayage piglets with negative Mhp antibodies,and randomly divided into control group,Chinese herbal medici-ne group,vaccine group,and Chinese herbal medicine+vaccine group,continuous feeding of No.3 Chinese herbal medicine for 10 days.Researching the influence of No.3 herbal additives on antibody levels of piglets against swine mycoplasma pneumonia vaccinated piglets,part of blood and blood biochemical parameters,IL-2,IL-4,IFN-γ content,T lymphocyte subsets and T lymphocyte proliferation.The results are as follows:1.The effect of No.3 of Chinese herbal medicine additive on antibody levels of mycoplasma pneumoniae of swine.To study the effect of No.3 Chinese herbal medicine additive to the inactivated vaccine immunization test, piglets of vaccine group and Chinese herbal medicine+vaccine group were vaccined the first Ann hectogram vaccine in the age of 15,and in the age of 30 were vaccined the second vaccine.Respectively gathering serum before the experiment, in the age of 30 and in 50 days of age, detecting the levels of swine mycoplasmal pneumonia antibody by adopting the method of ELISA. Results: in the age of 30 and 50 days of age, the S/P value of vaccine group and Chinese herbal medicine + vaccine group were higher than blank control group and Chinese herbal medicine(P<0.01);S/P value of Chinese herbal medicine+vaccine group is higher than the vaccine group(P<0.05).Display:No.3 Chinese herbal medicine additive can improve The level of piglets mycoplasma pneumonia inactivated vaccine immune antibody.2.The effect of No.3 Chinese herbal medicine additive to part of the piglets ’routine blood and blood biochemical indicators.To study the effect of No.3 Chinese herbal medicine additive to part of the piglets’ routine blood and blood biochemical indicators,the test gathered blood respectively in before the experiment,in the age of 30 and in 50 days of age,and adopts fully automatic blood cell analyzer and blood biochemical analyzer for testing.The result shows:when the age of 30 days,compared with the control group:Chinese herbal medicine, vaccine group,Chinese herbal medicine+vaccine group have the WBC increased by 4.75%,8.72% and 18.62%(P<0.05);LYM increased by 9.83%,15.08% and 25.57%(P<0.05);Gran increased by 15.12%, 21.00% and 36.12%;RBC increased by 8.36%,2.85% and 9.15%;TP increased by 15.81%(P<0.05), 6.67% and 16.85%(P<0.05);ALB increased by 6.37%,-0.74% and 10.61%;GLO increased by 38.71%(P<0.05),24.61%,32.00%(P<0.05).When the age of 50 days, compared with the control group:Chinese herbal medicine,vaccine group,Chinese herbal medicine+vaccine group have the WBC increased by 8.39%, 34.5 4%,36.45%;LYM increased by 28.80%,12.58% and 31.78%;Gran increased by 12.23%,0.53% and 29.25%; RBC increased by 11.45%,7.21% and 27.23%(P<0.05);TP increased by 14.87%(P<0.05), 12.30%(P<0.05) and 23.45%(P<0.05);ALB increased by 11.08%, 6.59% and 14.07%(P<0.05);GLO increased by 22.81%,24.27%,43.10%(P<0.05).It showed that the No.3 Chinese herbal medicine can significantly improve the number of piglet peripheral white blood cells,lymphocytes,red blood cells and the content of total protein,albumin and globulin in serum.3.The effect of No.3 Chinese herbal medicine on the content of IL-2,IL-4,IFN-γin piglets serum.To study the effect of No.3 of Chinese herbal medicine on the content of IL-2,IL-4,IFN-γ in piglets serum, whicht were tested by ELISA kits.the result shows:at the age of 30 days,compared with the blank control group:Chinese herbal medicine group,vaccine group,Chinese herbal medicine+vaccine group content of IL-2 increased by 0.92%,4.14% and 5.32%(P<0.05);content of IL-4 increased by 3.44% and 14.22%(P<0.05) and 12.18%(P<0.05);content of IFN-γ increased by 4.11%,6.55% and 21.12%(P<0.05).When 50 days of age,compared with the blank control group:Chinese herbal medicine group,the vaccine group,the Chinese herbal medicine+vaccine group,the content of IL-2 increased by 4.10%(P<0.05),1.05% and 12.75%(P<0.05);the content of IL-4 increased by 7.63%,7.42% and 8.54%;the content of IFN-γ increased by 4.15%,4.06% and 12.44%(P<0.05).Description:No.3 of Chinese herbal medicine additive improves the content of IL-2,IL-4,and IFN-γ piglets serum,especially improve the content of IL-2,IL-4,and IFN-γ piglets serum of the Chinese herbal medicine+vaccine group.4.The effect of No.3 Chinese herbal medicine additive on T lymphocyte subgroups and T lymphocyte proliferation of piglets in vitro.To study The effect of No.3 Chinese herbal medicine additive to T lymphocyte subgroup and the T lymphocyte proliferation of piglets in vitro, the test using flow cytometry test peripheral blood T lymphocyte subsets CD4~+,CD8~+ percentage change;The CCK-8 method is used for T lymphocyte proliferation test in vitro.Results showed:when 30 day age group,compared with control group:Chinese herbal medicine group,vaccine group,Chinese herbal medicine+vaccine group which percentage of CD4~+T cell increased by 8.96%, 16.91% and 26.24%;percentage of CD8~+ T cells increased by 3.33%,4.62% and 7.02%(P < 0.05);The value of CD4~+/CD8~+ rose by 6.88%,11.85% and 19.41%(P < 0.05);T lymphocyte stimulation index(SI) were increased by 5.47%,3.48% and 10.02%(P<0.05).When 50 days of age, compared with the control group:Chinese herbal medicine group, vaccine group, Chinese herbal medicine + vaccine group which the percentages of CD4~+T cell increased by 13.43%,10.47% and 33.30%(P<0.05); percentage of CD8~+T cells were increased by 2.42%,5.00% and-2.21%;The value of CD4~+/CD8~+ respectively raised 14.87%,6.49% and 32.68%(P < 0.05);T lymphocyte stimulation index(SI) were increased by 6.90%,2.24% and 9.80%(P<0.05).It showed that the No.3 Chinese herbal medicine additive can improve the percentage in peripheral blood CD4~+T cells,the the proportion of CD4~+/CD8~+ and T lymphocyte proliferation in vitro,especially can obviously improve the the percentage of CD4~+T cells and CD8~+T cells,the proportion of CD4~+/CD8~+ and the stimulation index(SI) of Chinese herbal medicine +vaccine group.Therefore,the No.3 Chinese herbal medicine can improve the pig mycoplasma inactivated vaccine antibo-dy titer,the number of white blood cells,red blood cells and lymphocytes in the blood,the content of total protein,albumin and globulin in serum and the content of IL-2,IL-4 and IFN-γ in serum,the percentage CD4~+T cells,the proportion of CD4~+/CD8~+and T lymphocyte proliferation in vitro,so as to improve the immunity and disease resistance of the piglets. |
