Rosa davurica Pall is a plant of Rosaceae, Rosoideae, deciduous shrubs. It is produced in the area of Changbai Mountain. Distributed both in the northeast and north of China, it is a folk medicine, with bleeding and broad-spectrum antimicrobial activity. It’s main uses are for the treatment of menstrual blood, dysfunctional uterine bleeding, chronic bronchitis, enteritis, dysentery, cystitis and nephritis. There are many reports both domestic and foreign, about its chemical constituents, but more common are the reports of anti-inflammatory and antioxidant activity. There are less studies about the chemical constituents and hepatoprotective activity of constituents. Therefore, the separation of its active ingredients, to protect the liver and to further define the material basis of its hepatoprotective activity, has important theoretical and practical significance.In this study, we separate the active ingredients to protect the liver of Rosa davurica Pall. Using various methods of column chromatography, including normal phase silica gel, RP-silica gel, Sephadex and other methods. Through the activity tracking, twelve compounds were isolated from the ethyl acetate extracts of Rosa davurica Pall, and further through the physical and chemical methods and spectral analysis (1H-NMR,13C-NMR, 1H-1H COSY, HSQC, etc.) identified the structure of 11 compounds. The structure of these compounds were:β-sitosterol- glucopyranoside (compound 1)、2α,3β,19α-trihydroxy -12 en -28 oic acid、(compound 2)、2α, 3a,19a-trihydroxy-12 en-28 oic acid glucoside (compound 3)、β-sitosterol (compound 4)、Betulinic acid(compound 5)、3β-hydroxy-12 en-28 oic acid (Compound 6)、2α,3α, 19a-trihydroxy-12 en-28 oic acid、(Compound 7)、3-β-D-glucose (4,5)-dihydroxy methyl ester (compound 8)、catechin(+)(compound 9)、2β,3α,19α,24α-tetrahydroxy-12 en-28 oic acid (compound 10)、1a,2β,3β,19α-tetrahydroxy-12 en-28 ursolic acid (compound 11).We investigated that 4 compounds isolated from Rosa davurica Pall which have the protective effects against t-BHP-induced HepG2 cell damage by MTT method. the test data of EC50 as follows:compound 10,44.8±0.16 uM; compound 1,6.43±0.11 uM; compound 3,7.45±0.22 uM; compound 6,12.6±0.27 uM.In conclusion, compound 10 have the protective effects against t-BHP-induced HepG2 cell damage, In addition, compound 8、compound 3、compound 6 have significant protective effects.This subject not only increases understanding of its chemical compositions and active ingredients to protect the liver, but also helps with further research and development of new liver protection provides the scientific basis of drug activity.
