The ripe fruit of Rosa davurica Pall.named Rosa davurica,is widely distributed in Northeast,Mongolia and Shanxi province.It was used in the food field,such as functional drinks,health care products,drugs and so on.Researches have shown that there are plenty of chemical components in fruit of Rosa davurica Pall.,for example:flavonoids,vitamin,saponins,polysaccharides,and so on.The pharmacology study states clearly that it has the functions of antioxidant,anti-aging,promoting digestion and antiradiation.The analysis and preparation of flavonoid aglycones,and separation of chemical components from Rosa davurica Pall.extracts were discussed in the paper,which may promote comprehensive utilization of active,rich and nature fruit sourcs.First,the content determination method of two kinds of flavonoid aglycones in the fruit extracts of Rose davurica Pall.with high performance liquid chromatography(HPLC)and methodology study were investigated.Second,applying the response surface method the preparation of flavonoid aglycones with cellulose was optimized on the result of single factor experiment.Finally,using semi-preparative liquid chromatography,column chromatography and recrystallization methods,four components were isolated from the extracts and their structure were confirmed by thin layer chromatography(TLC)and HPLC methods.The determination of quercetin and kaempferol in the fruit extracts of Rose davurica Pall.with HPLC at the same time was established.The standard curve of quercetin was lineared in the range of 0.0080~0.6000 ?g(r=0.9997)and the standard curve of kaempferol was lineared in the range of 0.0025~0.1872 ?g(r=0.9995);the average rate of quercetin was 103.92%(RSD=0.29%,n=6)and the average rate of kaempferol was 95.50%(RSD=0.73%,n=6).The HPLC method offered in the paperhas lots of advantages,such as easy operation,good reproducibility and high accuracy which can apply for quality control of flavonoid aglycones.Using fruit of Rosa davurica Pall.as material the enzymatic hydrolysis process of flavonoid glycosides was optimized by single factor and response surface applying peak area and hydrolysis rates as indexes for investigation.The results have shown that when the enzymolysis temperature was 60.2 ?,the enzymolysis time was 2.1 h,the enzymolysis pH was 3.2,the enzyme concentration was 20.0 mg/g and the solid-liquid ratio was 1:150 the hydrolysis rate of total flavonoids was higher than other conditions.Compared with the acid hydrolysis concidition,enzymplysis technology could not only low the reaction temperature but also improve the yield of flavonoid aglycones,which reflect great development value.Rutin,hyperin,quercetin and kaempferol were isolated from the fruit extracts of Rosa davurica Pall.by semi-preparative liquid chromatography,column chromategraphy and recrystallization methods.They were identified by TLC and HPLC,which provided a molecular basis for the comprehensive utilization of fruit extracts of Rosa davurica Pall..
