Objective To detect the expressions of miRNA-21and miRNA-205in maiden andrecurrence urothelial cell carcinoma of bladder and explore the relationship betweenmicrolRNAs(miRNAs) and urothelial cell carcinoma of bladder.Methods2-△△Ctfluorogenic quantitative reverse transcription PCR(FQ-PCR) method wasused for quantitative analysis of the expression of miRNA-21and miRNA-205in40maidenand recurrence urothelial cell carcinoma of bladder tissues.Results1.Compared with maiden tissues, the recurrence tissues showed significant miRNA-21over-expression (P<0.05).In the maiden tissues compared with non muscle-invasive bladdercancer, muscle-invasive bladder cancer showed significant miRNA-21over-expression(P<0.01),while in the recurrence tissues compared with non muscle-invasive bladder cancer,muscle-invasive bladder cancer showed significant miRNA-21over-expression also(P<0.05).2.Compared with maiden tissues, the recurrence tissues showed significant miRNA-205down-regulation (P<0.05).In the maiden tissues compared with non muscle-invasive bladdercancer, muscle-invasive bladder cancer showed significant miRNA-205down-regulation(P<0.01),while in the recurrence tissues compared with non muscle-invasive bladder cancer,muscle-invasive bladder cancer showed significant miRNA-205down-regulationtoo(P<0.05).3.Maiden cancer characterized as muscle-invasive bladder showed amiRNA-21:miRNA-205ratio at5.8-fold higher than the quantitative ratio obtained from nonmuscle-invasive bladder(P<0.01),while in the recurrence the ratio is5.2-fold(P<0.05). Conclusions1.Compared with maiden tissues, the recurrence tissues showed significant miRNA-21over-expression and miRNA-205down-regulation,speculated that they may be involved inbladder urothelial carcinoma recurrence.2.Compared with non muscle-invasive bladder cancer, muscle-invasive bladder cancershowed significant miRNA-21over-expression and miR-205down-regulation, speculated thatthey may have relation with muscle-invasive.3.Teach us to find the right drug suppression of microRNA expression, improve lowexpression of microRNA genes available treatment for bladder cancer at the molecular levelwith new target.The over expression of miRNA-21and lower expression of miRNA-205mayparticipate in recurrence urothelial cell carcinoma of bladder and have relationship withmuscle invasive.
