PART 1 The expression of miRNA-141 and miRNA-375 in the different tissue samples from prostateObjective:To explore the value of miRNA-141 and miRNA-375 in clinical diagnosis of prostate cancer through detecting the expression of miRNA-141 and miRNA-375 in the tissue samples from prostate cancer and benign prostatic hyperplasia and analyzing the intensity and the difference of expression.Methods:30 fresh prostate tissue samples were collected from hospitalized patients of Department of Urology in First Affiliated Hospital of Nanchang University from August,2010 to March,2011, among which 15 malignant tissue samples were from patients after Laparoscopic radical prostatectomy, and the other 15 tissues were from patients after transurethral resection of prostate(TURP). The clinical staging was learnt from reviewing and analyzing the clinical data of hospitalized patients. The expression of miRNA-141 and miRNA-375 in the tissue samples from Prostate cancer (PCA) and benign prostatic hyperplasia (BPH) was detected by RT-PCR. SPSS 12.0 software was used for statistical analysis of the experimental data to show the expression difference. All the data were in the form of "mean±standard deviation" and analyzed with one-way ANOVA. P value less than 0.05 was considered statically significant. the Spearman bivariate correlation analysis was used,P value less than 0.05 was considered statically significant.Results:The expression of miRNA-141 in the tissue samples from prostate cancer was 11.76±10.07 times respectively of that from benign prostatic hyperplasia, while the expression of miRNA-375 in the tissue samples from prostate cancer was 18.56±13.27 times respectively of that from benign prostatic hyperplasia, This experiment showed that the expression intensity of miRNA-141 and miRNA-375 is positively correlated with TPSA, Correlation Coefficient is 0.70 and 0.529, but have unrelated to Gleason grade.Conclusion: 1. The expression difference of miRNA-141 and miRNA-375 in prostate tissue suggests that miRNA-141 and miRNA-375 are likely to be significant tumor markers.2. Further researches show that the expression of miRNA-141 and miRNA-375 is related to pathological grade.PART 2 The expression of miRNA-141 and miRNA-375 in the serum samples from prostate cancer, benign prostatic hyperplasia and normal humanObjective:To explore the value of miRNA-141 and miRNA-375 in clinical diagnosis of prostate cancer through detecting the expression of miRNA-141 and miRNA-375 in the serum samples from prostate cancer, benign prostatic hyperplasia and normal human and analyzing the intensity and the difference of expression.Methods:327 serum samples were collected from hospitalized patients of Department of Urology in First Affiliated Hospital of Nanchang University from August,2010 to March,2011.20 samples of prostate cancer,20 samples of benign prostatic hyperplasia and 20 samples of normal human serum were selected according to the medical records. The expression difference of miRNA-141 and miRNA-375 in prostate tissue suggests that miRNA-141 and miRNA-375 are likely to be significant tumor markers.2.
