Objective:The aim of the study was to investigate the expression of interleukin-8(IL-8)、 interleukin-6(IL-6) and interferon-γ in normal prostate, benign prostatic hyperplasia by means of immuhistochemical S-P method. To evaluate the effect of inflammation on the development and progression of BPH, the relationship between these cytokines and symptom score was analysed.Material and Methods:Patient materialHistologically normal prostates obtained at autopsy from10men (18-35years old) without histories of reproductive or endocrine disease were used. The sever ones were obtained from prostate tissues derived from40patients undergoing transurethral resection of the prostate (TURP) for BPH. Patients were excluded if they had a history of prostate cancer, or there was suspicion of any urological cancer.ImmunohistochemistryImmunohistochemical S-P method was used on50tissues for the detection of IL-8、IL-6and IFN-y expression.Results:(1) In BPH specimens,80%(32of40) in the IL-8group expressed positive or strong positive. In the histologically normal prostates tissue, the positive rate of IL-8was20%(2of10). There were significant difference(P<0.05) when comparison was made between BPH specimens and the histologically normal prostates tissue.(2) In BPH specimens,77.5%(31of40) in the IL-6group expressed positive or strong positive. In the histologically normal prostates tissue, the positive rate of IL-6was20%(2 of10). There were significant difference (P<0.01) when comparison was made between BPH specimens and the histologically normal prostates tissue.(3) In BPH specimens,82.5%(33of40) in the IFN-γ group expressed positive or strong positive. In the histologically normal prostates tissue, the positive rate of IFN-γ was30%(3of10). There were significant difference (P <0.05) when comparison was made between BPH specimens and the histologically normal prostates tissue.(4) A positive correlation was observed between the expression of IFN-γ and IL-8and that of IL-6in hilar cholangiocarcinoma(P <0.05).Conclusions:Multi-factors were involved in the etiopathogenisis of BPH. IFN-γ is positively correlated to IL-6and IL-8, which demonstrates that inflammation plays an important role in BPH pathogenesis, and this provides a new target for the treatment of BPH.
