PINELLIAE RHIZOMA Origins from the dry tuber of Pinellia ternatat (Thunb.)Breit., with the effects of drying dampness to eliminate phlegm, calming the adverse-risingenergy to vomit, disintegrating abdominal lumps to eliminate stagnation. Using forexcessive phlegm, cough, dyspnea, headache, vomiting and regurgitating, and painfulabdominal mass. As one of the commonly toxicity Chinese medicines, popularly used theprocessed product. With the understanding of the long-standing irritant toxicity ofRHIZOMA PINELLIAE, there are many disputes on the toxicity of the material basis andmechanism. For the past few years, the researches of the pharmacology and chemicalconstituent are more and more deeply; have reported variety of pharmacologic actions andactive components, but what is the potency substance related to traditional efficiency stillhave not been resolved absolutely, resulted in indefinite positioning of the clinical potencyand imperfect quality evaluating standard.Firstly, this paper established a quality evaluation pattern which is based on greyrecognition. As based on the quantitation and comparative analysis of the activecomponents in PINELLIAE RHIZOMA, such as, organic acids, polysaccharides, alkaloidsand nucleosides. According to the Grey Incidence Degree numerical methods of GreyTheory, to establish Grey correlation model which is used to evaluate the quality of thePINELLIAE RHIZOMA. Provide a synthetic multi-component evaluating method underthe situation that the representative active components are indefinite.Secondly, in order to provide the most relevant active component as the most simplequality controlling marker, the correlation between active component and potency wasinvestigated by Grey correlation analysis model. Base on the study and analysis of the animal experiment on the antitussive and expectorant action of PINELLIAE RHIZOMA,PINELLIAE RHIZOMA PRAEPARATUM CUM ALUMINE, PINELLIAE RHIZOMAPRAEPARATUM, PINELLIAE RHIZOMA PRAEPARATUM CUM ZINGIBERE ETALUMINE, and then integrate with the organic acid, polysaccharide, alkaloids andnucleosides contents of the samplers. According to the factor analysis by Grey IncidenceDegree numerical methods of Grey Theory, reach the most relevant active component andprovide basis for PINELLIAE RHIZOMA quality evaluation and quality control indexesselection.The quantitation data of active components show that it is significantly differentamong different batches, and it is able to evaluate the quality of PINELLIAE RHIZOMAby Grey correlation model. Furthermore, the quantitation data of the processed samplesshow that the content of active components decreased to different degree except the contentof polysaccharide in PINELLIAE RHIZOMA PRAEPARATUM CUM ALUMINE. Thepharmacological experiment show that the mice cough incubation period prolongatedsignificantly by the4different samples (P<0.05or P<0.01), the PINELLIAE RHIZOMA,PINELLIAE RHIZOMA PRAEPARATUM, PINELLIAE RHIZOMA PRAEPARATUMCUM ZINGIBERE ET ALUMINE Significantly reduced the frequency of mice cough(P<0.05or P<0.01). And the PINELLIAE RHIZOMA is more effective than PINELLIAERHIZOMA PRAEPARATUM CUM ALUMINE at prolongating the mice cough incubationand reducing the mice cough, having the better potency. The PINELLIAE RHIZOMAPRAEPARATUM CUM ZINGIBERE ET ALUMINE can increase phenolsulfonphthaleinexcretion quantity of trachea in mice significantly (P<0.05), having the expectorant effect.The PINELLIAE RHIZOMA group with the larger quantity of phenolsulfonphthaleinexcretion is more effective than PINELLIAE RHIZOMA PRAEPARATUM CUMALUMINE group and PINELLIAE RHIZOMA PRAEPARATUM group. It’s seemed tobear out in a sense the saying that the raw PINELLIAE RHIZOMA decoction has bettereffect in eliminating phlegm and relieving cough. According to the correlation studybetween active component and potency based on the Grey correlation model, the sorting ofthe Grey Incidence Degree with regard to the mice cough incubation period is alkaloids> nucleosides>polysaccharides>organic acids; the sorting of the Grey Incidence Degreewith regard to the mice cough is alkaloids>nucleosides>organic acids>polysaccharides;the sorting of the Grey Incidence Degree with regard to the Tracheal phenol red excretion isalkaloids>nucleosides>polysaccharides>organic acids. So, the alkaloids are the mostrelevant component to potency, as a quality control marker to establish the quality controlstandard, at least.
