Study On Quality Standards And Organic Acids Of Rhizoma Arisaematis And Rhizoma Pinelliae Pedatisecta

Arisaematis Rhizoma,belonging to the Arisaema Mart.In Araceae,it is recorded to be tuber of Arisaema erubescens(Wall.)Schott.,Arisaema heterophyllum Blume.and Arisaema amurense Maxim.Its nature is warm,acor and spicy tasted.And in clinic,it has good for apoplexy,intractable phlegm,skew mouth and eyes,hemiplegia,numbness of hand and foot,dizziness caused by wind phlegm and so on..External has the effects of riding boils swollen,snake bite and so on.Rhizoma Pinelliae Pedatisecta is the tuber of Pinellia pedatisecta Schott,belonging to Pinellia Tenore in Araceae family.The nature of it is little smell,spicy taste,irritation of the tongue and warm;It links lung,liver and spleen,which takes effect in eliminating phlegm,tumescence and spasm.lt is first recorded in the "Shen Nong Ben Cao Jing",and it hasn't been recorded in "Chinese Pharmacopoeia".The small size one is often treated as Rhizoma Pinelliae,while most of them make up mainstream products of Rhizoma Arisaematis.The use has been disordered since ancient times.In clinic,it has good for apoplexy,intractable phlegm,skew mouth and eyes,hemiplegia,numbness of hand and foot,dizziness caused by wind phlegm and so on.In this thesis,In accordance with the edition of "Chinese Pharmacopoeia",doing some study of Standards of the cutting and drying technolog in the slice of Rhizoma Arisaematis and Rhizoma Pinelliae Pedatisecta.And studied on the characters,identification,inspection,extract,determination and so on.Therefore,the pharmacy effect test were done on polysaccharides in the organic acid sites.This paper includes the following parts.1.Literature reviewThe thesis made a systematic summary for Rhizoma Arisaematis and Rhizoma Pinelliae Pedatisecta on resource distribution,cultivar identification,chemical composition and determination of the content,pharmacological effects and clinical applications.lt is showed that it has a long history in the common toxic Chinese medicine.It has a confusion used since ancient times,and it must be further studied to find the effective composition.In addition,it has the advantage in antitumour,but its effective ingredients and mechanism is not clear.2.Study on Quality Standards of Pieces(1).Study on pieces of technologyThrough studiing of the extracting rate,the content of total flavonoids and the total organic acid as index,it is showed that the pieces which cutting process using crosscutting,then drying(2).Study on Quality StandardsDoing some study of Standards of 12 batches of Rhizoma Arisaematis and Rhizoma Pinelliae Pedatisecta,studing the characters,identification,inspection,extract,determination and so on,and then drafted the bill of the quality stardard.3.Study on organic acids(1).The study on organic acidsThe application of GC-MS to analysis the organic acids,and it indicated that 41 components were identified through searching the NIST2010 database,it contained twenty-nine monobasic acids,ninet dicarboxylic acids and three tribasic acids.Saturated fatty acid and unsaturated fatty acids accounted for about 63.49%and 36.51%of the total peak area.And the citric acid accounted for about 31.5%.(2)Determination of total organic acidsIt has been developed the method that by citric acid as index based on the formation of hydroximic acid from the organic acid in the presence of dlcyclohexylcarbodiimide(DCC)and hydroxylamine perchlorate(HAP)as well as the formation of purple ferric hydroxamate from hydroximic acid in the presence of acidic ferric perchlorate solution.Through the study methodology,approach was consistent with requirements determination:citric acid has a good linear relationship between the concentrations and the absorbance of schaftoside in the ranges of 0.078?0.390 mg/mL,r=0.9998,and the average recovery was 99.91%(RSD=1.32%,n=9).(3).Study on the separation of organic acidThrough the comparative study of different resin palm to the separation effects of organic acid in Rhizoma Arisaematis and Rhizoma Pinelliae Pedatisecta,the results showed that the application of HB16 and D315 anion exchange resin can effectively separate the organic acid.(4).Study on pharmacological activity inhibiting of cancer cellsIn vitro cytotoxicity of MTT method,the results showed that organic acids sites have significant anti lung cancer,gastric cancer and liver cancer,and through the comparison of different concentrations of drugs inhibiting rate of cancer cells,the inhibition and dose has the effect of positive correlation.And it has a significant role in improving NO in Rhizoma Arisaematis and Rhizoma Pinelliae Pedatisecta,there is a definite relation in its anti tumor.
