Objective:To explore the expression of iodide transporter related SLC5A5and SLC26A4gene in multinodular goiter, and analyze the expression and distribution of their coding protein NIS and Pendrin.Method:Specimens of thyroid tissue were obtained from40multinodular goiter patients undergoing surgery in Tianjin general hospital while control group were obtained from40normal thyroid tissue. Real time polymerase chain reaction(RT-PCR) was used to test SLC5A5gene and SLC26A4gene in two groups. Also, Western blot and immunohistochemistry was used to determine the expression and distribution of NIS and Pendrin protein.Result:1. The SLC5A5gene mRNA expression in multinodular tissue is significantly increased in comparison with normal nonodular goiter tissue. Difference between the two groups was significant (P=0.04). Also, SLC26A4gene mRNA expression in multinodular goiter tissue is significantly increased in comparison with normal nonodular tissue. Difference between the two groups was significant.2. The result of Western blot showed that the Pendrin protein expression in multinodular goiter group (1.6877±2.24157) was higher than in normal tissue (1.07256±0.7891), the difference was significant.3. The immunohistochemistry results showed that NIS positive ratio in multinodular goiter tissue and normal tissue was60%and37.5%respectively.14samples of24NIS positive patients with multinodular goiter were mainly expressed in the cytoplasm, the most samples in normal tissue were expressed in the membrane. The difference was significant(P<0.05). Also, there was positive expression of Pendrin in24patients (60%). And there was positive expression of Pendrin in14patients in normal tissue group. The difference was also significant(P=0.025).15samples of24Pendrin positive patients with multinodular goiter were mainly expressed in the cytoplasm.Conclusion:The expression of SLC5A5mRNA is increased in multinodular goiter tissue. Also, SLC26A4mRNA and its coding protein Pendrin expression are increased. The NIS protein and Pendrin protein in multinodular goiter were mostly expressed in the cytoplasm rather than in the functional membrane. It implied that iodide transporter function maybe damaged when multinodular goiter occurs.
