Gut immunity is an important part of the Drosophila’s innate immune systems. To observe the effect of the Saussurea laniceps (S. laniceps) extracts on gut immunity of Drosophila melanogaster. D. melanogaster treated with pathogenic microorganisms, inflammatory irons and metal iron with or without S. laniceps extracts. Their survival rates, intestinal epithelial cell death, melanoma formation and the intestinal morphology changes were observed. S. laniceps extracts could help protect D. melanogaster from Beauveria bassiana infection and attenuate the inflammatory damage caused by SDS, NaCl or KA1(SO4)2. The survival rate of D. melanogaster was greatly increased and intestinal epithelium cell death was significantly reduced and intestinal wall was intact. These results demonstrate that S. laniceps extracts could enhance the resistance of D. melanogaster to fungal infection and inflammatory injury.Rhodiola crenulata (R. crenulatd) is a traditional folk medicine in China. R. crenulata extracts possess multiple pharmacological activities, such as anti-oxidant activities, anti-high altitude illness activity and neuroprotective effects. To study the protective effect of R. crenulata extracts on gut immunity of D. melanogaster. Wild type flies fed standard cornmeal-yeast medium were used as controls. Experimental groups were supplemented with2.5%R. crenulata aqueous extracts in standard medium. Survival rate was determined by feeding pathogenic microorganisms and toxic compounds. The levels of reactive oxygen species (ROS) and dead cells were detected by dihydroethidium (DHE) and7-amino-actinomycin D (7-AAD) staining, respectively. The expression of antimicrobial peptides was evaluated by RT-PCR, and morphological change of the intestine was imaged by an Axioskop2Plus microscope. The R. crenulata extracts can increase the survival rates of adult flies and levels of antimicrobial peptide genes after pathogen or toxic compound ingestion. Moreover, decrease the levels of ROS and epithelial cell death, resulting in improved intestinal morphology. The pharmacological action of R. crenulata extracts from Tibet was greater than that from Sichuan. These results demonstrate that R. crenulata extracts could significantly improve the gut immunity of D. melanogaster.
