[Background]Invasion and metastasis of Cancer is the main cause of death in patients, however, tumor invasion and metastasis is a complex process of multiple genes involved in a multi-step, wherein the cell adhesion, cell migration and matrix dissolution.there are the three steps of invasion and metastasis of cancer. Recent studies show that CD44v6 and MMP-9 involved in the development of tumors, especially in lymph node metastasis and invasive growth, they also found their expressions may closely related with the prognosis.[Objective]:The study aims to detect the expression of MMP-9 and CD44v6in the gastric mucosa tissues of 60 normal cases and the adjacent non-cancer tissues of 60 cases by double immunohistochemical staining technique. And collect there clinical data at the same time. Finally we analyzed the expressions of MMP-9 and CD44v6 and the roles in metastasis of tumor invasion both in the process and relevance.[Method]Collect the pathological data and clinical data integrity of 60 cases treated from March 2013 to March 2014 period and have accepted gastric cancer surgery in the First Affiliated Hospital of Zhengzhou University, select the adjacent non-cancer tissues (away 5cm from cancer tissues) of 60 cases among them, all patients were not received prior radiotherapy and chemotherapy, Chinese medicine and immunotherapy before surgery. Using double immunohistochemical staining to detect the expression of CD44v6 and MMP-9 in gastric cancer, the x2 test was performed for data analysis to comparing the relationship between metastasis of tumor invasion and there expression,and using Spearman correlation test to analysis tissue expression relationship between CD44v6 and MMP-9 in gastric cancer. All statistical analyzes were performed using the statistical software on the computer and apply SPSS 17.0 software package to carry out statistical treatment.[Result]:1.A positive expression of CD44v6 in gastric carcinoma was 81.67%, significantly higher than the rate of positive expression in adjacent normal gastric tissues 20.00%,there is a significant difference between the two (X2= 53.393 p<0.01).2. MMP-9 expression in gastric carcinoma was 73.33%, significantly higher expression rate in adjacent normal gastric mucosa 5.00%, there is a significant difference (X2=10.568 p<0.01) between the two.3. The results of x2test showed that the expression of CD44v6were significantly correlated (x2=9.013and 8.104 p 0.05) with tumor invasion and TNM stage. And we found no correlation (p> 0.05)with the number of lymph node metastasis, the gender, age, size and there site of occurrence.4.There was no significant correlation (p> 0.05) between MMP-9 expression and the number of lymph node metastasis, the gender, age, size and the site of occurrence in patients; and the expression of MMP-9 was significantly correlated (X2=9.882 and 8.304 p<0.05) with the depth of invasion and TNM stage.5. The expression of MMP-9 in gastric carcinoma and CD44v6 has some correlation (r= 0.336, P= 0.00<0.01), both of there expression was positively correlated in gastric cancer.[Conclusion]:1. Overexpression phenomenon of CD44v6 and MMP-9 exists in gastric cancer, it may play an important role in the progression of gastric cancer.2. The overexpression ofCD44v6 and MMP-9 in gastric carcinoma was significantly associated with tumor invasion and TNM stage.it may play a role in gastric tumor invasion and metastasis.3. The expression of CD44v6 and MMP-9 expression in Gastric carcinoma has correlated positively. both may play synergistic role in gastric tumor invasion and metastasis.
