Objective To observe the differences of endocardial pressure and heart rate of rat with tachycardia or bradycardia and rat with P receptor knockout, to explore the two-way regulation effect of cardiac function by acupuncturing Lower He-sea point, Front-Mu Point, Back-Shu Point and Yuan-Source Point in pericardium meridian and their matching acupoints, and to discuss whether the two-way regulation effect is closely related to the functional stasis of preceptor or not.Methods Left ventricular endocardial pressure and heart rate were recorded after performing intubation tube through left ventricle so as to observe the effect of electro-acupuncture on rat cardiac function by acupuncturing Daling, Quze, Jueyinshu, Tanzhong. This study was divided into three parts including:1. Observe the effect of single Lower He-sea point (Quze), Front-Mu Point (Tanzhong),Back-Shu Point (Jueyinshu) and Yuan-Source Point (Daling) on ventricle endicardial pressure of normal rat and analyze the effective direction (rising or lowering) and effective quantities of single acupiont.2. Observe Observe the effect of matching acupionts of Lower He-sea point (Quze), Front-Mu Point (Tanzhong),Back-Shu Point (Jueyinshu) and Yuan-Source Point (Daling) on ventricle endicardial pressure of normal rat and analyze the effective direction (rising or lowering) and effective quantities of matching acupoints.3. Observe the effect of electro-acupuncturing single Lower He-sea point (Quze), Front-Mu Point (Tanzhong),Back-Shu Point (Jueyinshu) or Yuan-Source Point (Daling) on ventricular endocardial pressure of normal wild-type rat with Preceptor knockout and analyze the effective direction (rising/lowering) and effective quantities of single acupoint. Endocardial pressure and heart rate were considered as indexes and immediate effect within2min and post-acupuncture effect2min after removing needles were observed.Results (1) single acupoint:acupuncture change rate mean was greater than1%in immediate effect within2min and post-acupuncture effect2min after removing needles of electro-acupuncturing Daling and post-acupuncture effect2min after removing needles of electro-acupuncturing Quze and Jueyinshu. Daling showed obvious exciting effect while Quze and Jueyinshu presented slight exciting effect, acupuncture change rate mean of Daing, Quze and Jueyinshu was lower than1%under action of propranolol and isoproterenol, which showed no markedly changes. The effective direction of Quze mainly showed exciting while Jueyingshu was unstable. Acupuncture change rate mean of eclectro-acupuncturing Tanzhong was less than1%, showing no obvious changes, but the effective direction mainly showed inhibition.(2) The effect of matching acupionts:as for the effective direction under normal state,daling-tangzhong, jueyingshu-tangzhong,daling-quze mainly showed excitement state in immediate effect and post-acupuncture effect;quze-jueyinshu showed excitement state in post-acupuncture effect; quze-tangzhong, and daling-jueyinshu in immediate effect and post-acupuncture effect, andquze-jueyinshu in immediate effect mainly presented inhibition state. Under the state of propranolol, daling-quze, anddaling-jueyinshu in immediate effect and post-acupuncture effect, and daling-tanzhong in immediate effect showed excitement state; quze-jueyinshu, and quze-tanzhong in immediate effect and post-acupuncture effect,daling-tanzhong, andjueyinshu-tanzhong in post-acupuncture showed inhibition state. Under the state of isoproterenol, the effective direction ofquze-jueyinshu showed excitement state; the effective direction ofdaling-tanzhong, daling-quze,quze-tanzhong, anddaling-jueyinshu showed inhibition state.Under normal state, acupuncture change rete mean of onlyjueyinshu-tanzhong in post-acupuncture effect was greater than1%, showing that electro-acupuncture on jueyinshu-tanzhong had exciting effect on cardiac function in rat, while others were lower than1%showing no obvious changes. Under the state of propranolol, acupuncture change rete mean of only quze-jueyinshu in immediate effect and daling-quze in post-acupuncture effect was greater than1%, showing electro-acupuncture on quze-jueyinshu had inhibiting effect on cardiac function in rat, while electro-acupuncture on daling-quze had exciting effect on cardiac function in rat and others were less than1%which showed no obvious changes. Under the state of isoproterenol, the acupuncture change rate mean of quze-tanzhong in immediate effect and post-acupuncture effect, quze-jueyinshu and daling-jueyinshu in immediate effect were greater than1%, showing electro-acupuncture on quze-jueyinshu had exciting effect on cardiac function in rat, electro-acupuncture on quze-tanzhong and daling-jueyinshu had obvious inhibiting effect on cardiac function in rat, but others were lower than1%, showing no obvious changes.(3) Under the state of Preceptor knockout:the effect of electro-acupuncture on rat endocardial pressure:as for the effective direction, daling-quze in immediate effect and post-acupuncture effect mainly showed excitement stste; tanzhong-jueyinshu in immediate effect manly showed inhabitation state, but jueyinshu in post-acupuncture showed excitement state while tanzhong mainly showed inhibion state. The acupuncture change mean of quze in immediate effect and tanzhong in post-acupuncture effect was less than1%, showing no obvious changes.The acupuncture change mean of daling in immediate effect and post-acupuncture effect, and quze in post-acupuncture effect were greater than1%, showing that electro-acupuncture on daling and quze had exciting effect on cardiac function in rat. The acupuncture change mean of jueyinshu and tanzhong in immediate effect was greater than1%, showing that eletro-acupuncture on jueyinshu and tanzhong had inhibiting effect on cardiac function in rat.The effect of lectro-acupuncture on rat heart rate:as for the effective direction, daling in immediate effect mainly presented exciting state, but daling in post-acupuncture effect, and tanzhong in immediate effect and post-acupuncture effect showed inhibiting state. The acupuncture change rate mean of daling, quze and tanzhong in immediate effect and post-acupuncture effect was lower than1%, showing no obvious changes. The acupuncture change rate mean of only jueyinshu in immediate effect and post-acupuncture effect was greater than1%, showing that electro-acupuncture on jueyinshu had obvious inhibiting effect on cardiac function in rat.Conclusion:1. Single acupoint:Under normal state, electro-acupuncture on daling had exciting effect on cardiac in rat.2. Matching acupoints:Under normal state, electro-acupuncture on jueyinshu-tanzhong had exciting effect on cardiac function in rat. Under the state of propranolol, electro-acupuncture on quze-jueyinshu had inhibiting effect on cardiac function in rat while electro-acupuncture on daling-quze had exciting effect on cardiac function in rat. Under the state of isoproterenol, electro-acupuncture on quze-jueyinshu had exciting effect on cardiac function in rat while electro-acupuncture on quze-tanzhong and daling-jueyinshu had inhibiting effect on cardiac function in rat.3. The state of p*receptor knockout:The effect of electro-acupuncture on rat endocardial pressure:electro-acupuncture on daling and quze had exciting effect on cardiac function in rat while electro-acupuncture on jueyinshu and tanzhong had inhibiting effect on cardiac function in rat.The effect of electro-acupuncture on rat heart rate:electro-acupuncture on jueyinshu hadinhibiting effect on cardiac function in rat.The consolidated, it can be seen:acupuncture in different states, different parts of the acupoints, acupoints single bi-directional regulation effect is not obvious, p-adrenergic receptor knockout still have more obvious effects of acupuncture in mice after EA.
